Objectives To investigate the toxicity of hyperhomocysteinemia (Hhcy) on cerebrovascular endothelial cell of rats, and to provide experimental evidences for the new risk factor of ischemic cerebrovascular disease.Methods We fed male Wistar rats with a high methionine(Met)diet (2%)for 8 weeks, to establish the Hhcy rat model. The plasma total homocysteine(tHcy)levels were assayed by high-performance liquid chromatography method(HPLC);the concentration of plasma malon-dialdehyde(MDA)were measured by thiobarbituric acid reactive substance(TBARS)method, and the levels of plasma nitric oxide(NO)were analyzed by nitric acid reductase method .The analysis on rats middle cerebral artery and cerebral cortex were performed with transmission electron microscope(JEOL 100CX).Results1. At the beginning, the concentration of plasma tHcy were 13. 79 ± 2.66 μ mol/L. After 4 weeks, we successfully induced Hhcy model in rats by high Met diet. At the end of 4wk and 8wk, the plasma tHcy levels of model group were 22. 56 ± 3.10 μmol/L and 28. 83 ± 5.87 μ mol/L respectively, higher than those of the normal group (14.10±1.88μmol/L and 13.94 ± 2.76...
