Objective To identify the penumbra of variant ischemia andreperfusion period and the effect of the blocker of gapjunction-octanal on the penumbra.Methods Total 60 male Wistar rats were randomly divided into thefollowing groups: sham-operated group, contral:I6hR0.5h (ischemiafor 6 hours and reperfusion for 0.5 hour),I6hR1h,I6hR3h,I6hR6h,I6hR12h,I6hR24h,I6hR72h, octanol-treatment group(I6hR6h,I6hR12h),DMSO group. There were 6 rats in each group. Rats inexperimental groups were suffered focal cerebral ischemia-reperfusion through a nylon suture method. Praxiology changes andneurologic impairment were evaluated by Longa five-grade scoringstandard. The brains were stained with toluidine blue, cresyl viollet.The expression of Cx43 was detected by immuno-histochemistry. Thearea of the penumbra was measured and analyzed with an image analysissystem.Result All the animals of groups except sham-operated one displayedobvious manifestation of neurologic impairment. There was nodifference between each groups. The two methods correlated with eachother in measuring the penumbra. There was no difference of Cx43levels between each groups.
