Objective: To investigate the characters of hexokinase II gene sequence, and its polymorphisms and genetic susceptibility to Primary Hepatocellular Carcinoma(PHC) in Guangxi Chuang and Han people.Methods: Polymorphisms of HK-II gene were analyzed in 80 patients suffering PHC and 122 health controls.The partial sequence of HK-II gene was scanned by molecular and genetic techniques, including PCR, PCR-SSCP, PCR-RFLP , direct sequencing of PCR products.Results: (1) The mutation of CAA — CAT were found in exon 4 of HK-II gene, and the frequency was significantly higher in patients than in control groups(0.363 vs 0.172, P<0.05);(2) Mutation were also found in exon 14 of HK-II gene, but the frequency did not show obvious difference between cases and controls(0.0625 vs 0.0328, P>0.05); (3) The nucleotide substitutions of CCC-CCG were found in exon 15 of HK-II gene, and the total mutation frequency did not show apparent difference between patients and controls(0.675 vs 0.738, P>0.05); (4) No mutation was found in the region of promoter of HK-II gene either in patients with PHC or control subjects. (5)If the exon 4 of HK-II gene is imitative and the genotype is SS type at the same time, it is obvious difference between cases and controls(P=0.009).
