Acanthus ilicifolius L. (Acanthaceae), an important mangrove plant, has been embodied in Zhong yao da ci dian for its efficacy as an antipyretic, an expectorant, a detoxicant, and an analgesic. Folk medicine indications include hepatitis, asthma, rheumatism, paralysis, and snakebite. Analgesic and anti-inflammatory activity serves as the fundament of folk medicine treatment. Although chemical constituents from Acanthus ilicifolius and some of their pharmacological activity were investigated deep in recent years, we paid our study interest in exploring chemical constituents and antioxidant of Acanthus ilicifolius that were collected from Sankou, Hepu, Guangxi, China. As a result, eleven compounds were isolated by silica gel column chromatography, Vacuum Liquid Chromatography(VLC) , centrifugal thin layer chromatography(CTLC) and AB-8 Resin Adsorbing Method. On the basis of their chemical and physical properties, and spectral data such as IR, 1H-NMR, 13C-NMR, and ESI-MS, eight of them were structurally elucidated as Hexacosanoic acid(AI-l), Stigmasterol (AI-2), 2-Benzoxazolinone(AI-3), Stigmasterol-3-0- β -D-glucopyranoside (AI4), vanillic acid(AI-5), 4hydroxy-2- Benzoxazolinone (AI-6), Tetratriacontanol (AI-9), Quercetin (AI-11). Compounds AI-1, 5, 6, and 9 were reported first time for their being isolated from this mangrove species. The antioxidant , evaluated as per the ability to scavenge H2O2. was detected for different extracts from both stem and root of this plant by ECL that involves the establishment of H2O2-Luminol-CTMAB system. Detections showed that the ethyl acetate extract appeared strong activity of anti- oxidation, especialy the fraction 456-495. The active constituents may be of flavones, phenols, anthraquinones, or alkaloids. The effect of crud polysaccharids on antioxidant is not significant. Lipid peroxide (LPO ) in rat liver was detected by malondiadehyde(MDA) colorimetry. The crud polysaccharids has some extand inhi bitory effection on LPO.
