Objective To investigate the probable mechanism of normal cells had carcinomatous change induced by the supernatant of tumor cells, and to afford valuable cell models for looking for specific markers for clinical diagnosis, treatment, estimation of prognosis and developing anti-cancer drug in the future. Methods Through inducing the normal mouse fibroblast L929 cell line with the supernatant of mouse hepatocarcinoma H22 cell line, we made them having a carcinomatous change, then the induced cell line growing stablely was named L929-H22. By flow cytometry, immunocytochemistry, immunohistochemistry and cell enzyme-linked immunosorbent assay etc, we determined some characteristics and the expression of some proteins. Then we transplanted L929-H22 and L929 cells in nude mouse subcutis and observed if they had the potential of tumorigenesis. Result The cell cycle distribution of L929-H22 was markedly different with that of L929. As compared with L929 cell line, the proportion of L929-H22 cells in S-phase increased while in G0/G1-phase decreased significantly and the proliferating index hightened. The expression of MTP53,C-Myc,CD1,NF-κB,iNOS,VEGF,KDR,IGF-Ⅱ,IGF-ⅡR,EGFR,Survivin,CathepsinD was increased obviously, and the expression of P27,Bax,Fas,Caspase-3 ,E-cad ,nm-23 was decreased markedly. H22 cells expressed VEGF,IGF-Ⅱ. After transplanting L929-H22 cells and L929 cells in nude mice body, the nude mouse transplanting L929-H22 cells had transplant tumor, while the nude mouse transplanting L929 cells had no. By immunohistochemistry, we detected vimentin(+), S-100(-),Desmin(-),CK(-) and found the transplant tumor deriving from fibroblasts. So the induced cells L929-H22 had the potential of tumorigenisis. Conclusion 1. L929-H22 cell line induced with cultured supernatant of tumor cells H22 proliferate more actively than L929 cell line. 2. Induced cells L929-H22 apoptosis disability. 3. Induced cells L929-H22 have great potential of invasion . 4. Induced cells L929-H22 occure malignant conversion and may lead to tumorigenesis. 5.
