In this study a plasmid called pERE-luc which contained the human estrogen reponse element was constructed. Transfected the plamid into MCF7 cell transietly,stimulate the cells with E2 as positive control,test the luciferase activity to estimate the hormonal activity. Reporter gene-based in vitro recombinant cell assays for detection of environmental (anti)estrogenic endocrine disruptors were established; two plasmids called pSVARO and pMMTV-Luc was constructed, transfected the plamids into CHO cell transietly,stimulate the cells with DHT as positive control,test the luciferase activity to estimate the hormonal activity. Reporter gene-based in vitro recombinant cell assays for detection of environmental (anti)androgenic endocrine disruptors were developed; 16 chemicals were assessed for (anti)estrogenic activity and (anti)androgenic activity using the assays established . The results suggested the assays developed in the study were good at specificity and validity,they could serve as promising components of an in vitro strategy to assess the effects of chemicals,they could also provide information regarding the potential mechanism of action.
