Objective: In order to study the protective mechanism of the recombinant adenovirus carrying human IL-10 gene protect from hepatic hypothermic ischemia-reperfusion in rats. Method: we establish the model of orthotopic liver transplantation. In 3 groups. Having transfected for 72 hours, three groups were transplanted respectively, hIL-10 was checked with ELISA. EGFP was observed under the fluoroscopy, expression of mRNA of hIL-10, rTNF-α mRNA and rIL-1βmRNA was assayed by RT-PCR. Western blot for IκBα protein. EMSA for NF-κB activity. Result:we found that the rats pretreated with Ad-ILIO-EGFP expressed hIL-10 in livers and circulation at a high level. And NF-kB activity,TNF-α and IL-1β in liver and circulation are all decreased,Conclusion: hIL-10 could attenuated hepatic hypothermic ischemia-reperfusion.
