Objective To observe the effects of hirudin on proliferation and hyporplasia of extracellular matrix in glomerular mesangial cells stimulated by macrophage activated by ox-LDL.Methods Mesangial cell (MC) were co-cultured with macrophage normal and activated by ox-LDL from rats' abdominal cavity, and their medium after cultured 24h. At the same time, cells were co-cultured with hirudin(2.5U/ml, 5.0U/ml, 10U/ml). Its proliferation was measured by methyl thiazolyl tetrazolium (MTT) and flow cytometry metuy (FCM). RT-PCR was used to examining the expression of FN mRNA.Results â‘ macrophage, macrophage activated by ox-LDL and their medium all could promote the proliferation of MC .The function of macrophage activated by ox-LDL was the most. â‘¡ Activated macrophage could significantly up-regulate the expression of FNmRNA in MC. â‘¢Hirudin couldn't inhibit the proliferation of MC stimulated with macrophage activated by ox-LDL, but it could significantly down-regulate the expression of FNmRNA in MC, which was equal to heparin.Conclusion The effect of hirudin on inhibiting the proliferation of MC wasn't significant. But it could significantly down- regulate the expression of FN mRNA in MC. The results pointed out that hirudin may independently inhibit the hyperplasia of extracellular matrix in glomerular mesangial cells.
