[Objective]To investigate the relationship with the genotype and the allele frequency of (aldehyde dehydrogenase 2, ALDH2) and Alcoholic Liver disease (ALD) in Korean men and Han men of Yanbian area in China.[Methods]Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was used to detect ALDH2 single nucleotide Gene Polymorphism in the health control group (Korean 30, Han 20), alcohol dependent group (Korean 30, Han 15) and ALD group (Korean 30, Han 18) in Korean men and Han men of Yanbian area, we calculated and compared the genotype and the allele frequency in three groups .[Results]In Korean men the genotype frequency of ALDH2*1/1 between alcohol dependent group (83.3%) and ALD group (90%) was significantly higher than that in health control group (36.7% ) (P<0.05), but no significant difference was found between alcohol dependent group (83.3%) and ALD group (90%) (P>0.05); the genotype frequency of ALDH2*l/2 between alcohol dependent group (16.7%) and ALD group (10%) was significantly lower than that in health control group (53.3%) (P<0.05), but no significant difference was found between alcohol dependent group (16.7%) and ALD group (10%) (P>0.05); the genotype frequency of ALDH2*2/2 between alcohol dependent group and ALD group was 0%, the genotype frequency of ALDH2*2/2 in health control group was 10%, the difference of thegenotype frequency of ALDH2*2/2 in these three groups had statistic significance (P<0.05).In Han men the genotype frequency of ALDH2*1/1 between alcohol dependent group (86.7%) and ALD group (88.9%) was significantly higher than that in health control group (40%) (P<0.05), but no significant difference was found between alcohol dependent group (86.7%) and ALD group (88.9%) (P>0.05) ; the genotype frequency of ALDH2*l/2 between alcohol dependent group (13.3%) and ALD group (11.1%) was significantly lower than that in health control group (50%) (P<0.05), but no significant difference was found between alcohol dependent group (13.3%) and ALD group (11.1%) (P>0.05); the genotype frequency of ALDH2*2/2 between alcohol dependent group and ALD group was 0%, the genotype frequency of ALDH2*2/2 in health control group was 10%, the difference of the genotype frequency of ALDH2*2/2 in these three groups had statistic significance (P<0.05).In Korean men the allele frequency of ALDH2*1 between alcohol dependent group (91.65%) and ALD group (95%) was significantly higher than that in health control group (63.35%) (P<0.05), but no significant difference was found between alcohol dependent group (91.65%) and ALD group (95%) {P>0.05); the allele frequency of ALDH2*2 between alcohol dependent group (8.35%) and ALD group (5%) was significantly lower than that in health control group (36.65%) (P<0.05), but no significant difference was found between alcohol dependent group (8.35%) and ALD group (5%) (P>0.05).In Han men the allele frequency of ALDH2*1 between alcohol dependent group (93.3%) and ALD group (94.4%) was significantly higher than that in health control group (65%) (P<0.05), but no significant difference was found between alcohol dependent group (93.3%) and ALD group (94.4%) (P>0.05) ; the allele frequency of ALDH2*2 between alcohol dependent group (6.7%) and ALD group (5.6%) wassignificantly lower than that in health control group (35%) (P<0.05), but no significant difference was found between alcohol dependent group (6.7%) and ALD group (5.6%) (P>0.05).The genotype and the allele frequency of ALDH2 had no difference between Korean health control group and Han health control group. The genotype and the allele frequency of ALDH2 had no difference between Korean alcohol dependent group and Han alcohol dependent group (P>0.05). The genotype and the allele frequency of ALDH2 had no difference between Korean ALD group and Han ALD group (P>0.05).[Conclusion]There was no difference of frequencies distribution of ALDH2 gene polymorphism between Korean men and Han men of Yanbian area in China. ALDH2*2 was negative risk factors causing alcohol dependent and ALD.
