Objective:1.To evaluate the neuroprotective Effect of Cinepazide Maleate againstcerebral ischemia injury in rats . 2.To evaluate the possibility adenosinic-potentiating mechanism ofCinepazide Maleate against cerebral ischemia injury in rats .Methods:1. MCAO model in rate were prepared,114 SD rats were randomized into sham-operated group,control group and Cinepazide Moleate treated-group.2.the neurological dysfunction score(NDS) were assessed.3.Pathological changes were observed by light microscope and electromicroscope. 4.TTC staining were used to evaluate the infarct volume .5.Adopting immunohistochemistry method to detect the level of neuron-specific enolase (NSE) and glial fibrillary acidic protein (GFAP).6. Using TUNEL to label the condition of nenronal apoptosis 7.Highperformance liquid Chromatography (HPLC) technique was used to detect continuously the concentrations of Adenosine in the the three group respectively in brain after focal cerebral Ischemia reperfusion injury in ratsResults:Compared with those.in the control groups: The NDS were significantly increasing (P<0.05 ) 2.The infarct volume was significantly smaller(PO.05) 3.The Pathological changes were more minimal 4. the level of NSE expression significantly increased around infarction (PO.05) 5. the levels of the neuronal apoptosis decreased significantly (PO.05) .6 .GFAP expression in brain tissue around infarction area decreased significantly at 22h after reperfusion but increased significantly at 70h after reperfusion(PO.05) in the Cinepazide Moleate- treated groupThe Adenosine concentrations were significantly increased in the control groups than in the sham-operated group after cerebral ischemia and after reperfusion injury in rats (PO.05) ,Compared with those in the control groups, the concentrations of Adenosine after cerebral ischemia 20min.60min were significantly increased in the Cinepazide Moleate-treated group( P<0.05 ) .Although there were no statistical significant difference( P>0.05 ) , the Adenosine concentrations after cerebral reperfusion 15min.60min were slightly increased than these in the control groups.Conclusion:The concentrations of Adenosine sharply increased in the control group after focal cerebral ischemia in rats,but it only remain a short time.Cinepazide Moleate had significant protective effects to neuron around infarction areaand against cerebral Ischemia reperfusion injury in rats and improve the prognosis, it might be related to its protentiated endogenous protective effect of Adenosine by increasing the concentrations of endogenous Adenosine in brain.
