Objective: To investigate the role of pancreatic HGF and spleen TGF-β1on the proliferation and differentiation of hepatic oval cells in rats. Toexplore the regulation mechanism of endocrine manner on hepatic ovalcells.Methods: Ninety male Wistar rats weighing 150-200g, were randomlydivided into three groups: control group , 2-AAF/PH group( the modelof hepatic oval cells proliferation ) ,2-AAF/PH+SPL group(2-AAF/PH+Spleenctomy). establish the rat model2-AAF/PH (forhepatic oval cell proliferation),and the model of 2-AAF/PH+SPL.Observe the expression of HGF ,HGFmRNA in pancreas and ofTGF-β1,TGF-β1 mRNA in spleen after partial hepatectomy(PH) byimmunohistochemical and RT-PCR in the three groups.Results: 1,Proliferation of hepatic oval cells in three groups: there isnot any hepatic oval cells in control; in 2-AAF/PH, hepatic oval cellsoccurs at 4 days(9.23±4.82, n=5), increase to peak at 8 day(31.72±2.11,n=5), afterwards decrease, at 16d(15.80±6.45, n=5);In 2-AAF/PH+SPLgroup, hepatic oval cells became more numberous at 4 day(11.72±0.4,n=5), increase to peak at 8 day (39.23±0.18,n=5) and 12 day(37.08±1.87,n=5), and then begin decrease . 2,The express of HGFin pancreas and liver: In control group, pancreas tissue stain HGF weakly;In 2-AAF/PH group, the stain of HGF strongly, at 4 day ( 9.4±1.5,n=5),increase to peak at 8 day(21.5±7.2, n=5),and then decrease to normallevel at 16d(2.5±0.6, n=5).control vs 2-AAF/PH,P<0.05。Inaddition ,the trend of expression of pancreatic HGF had resemble to itsof liver. In 2-AAF/PH ,pancreas highly express the HGFmRNA, andIncrease vs control group, P<0.05.3,The express of TGF-β1 in spleenand liver: In control spleen, the macrophage in red pulps stain was weakly,at 8d (3.56±1.09, n=5);In 2-AAF/PH spleen, the express of TGF-β1increase with time, at 4d (7.14±1.05,n=5) and increased to peak value at8d (12.55±0.20, n=5) respectively. 2-AAF/PH vs control, P<0.05.Furthermore, 2-AAF/PH+SPL group compare with 2-AAF/PH, theexpress of TGF-β1 decrease distinctly,P<0.05.Conclusion: 1.HGF of pancreas may contribute to proliferation ofhepatic oval cells in rats. 2. TGF-β1 released from the spleen exerts agrowth inhibitory effect on hepatic oval cells in rats. 3.Pancreas andspleen exert particular regulation on different phases of liver regenerationby hepatic oval cells, through some cytokine. 4.There may be anendocrine manner on proliferation,differentiation of hepatic oval cells inrats.
