Objective: Basic fibroblast growth factor ( bFGF ) is an angiogenic factor that promotes proliferation endothelial cells,To investigate the effect of exogenous bFGF of different concentration on cultured human orbit's microvascular endothelial cells,Providing experimentation data for application bFGF to enhance angiogenesis of hydroxyapatite orbit implants. Methods:To obtain human orbits tissue casted away from surgery. The endothelial cells can be isolated and culturec by the method of twice filtration by various diameter stainless steel net and digestion by collagenase â…¡,The cells were identified by morphologic and immunohistochemical assays bFGF of different concentration 20ng/ml. 40ng/ml. 80ng/ml. 160ng/ml as experimental group was added in cultures,The cultures containing no bFGF served as control group .The number of endothelial cells was counted by MTT assay at 24h,48 h,72 h respectively. Results: Vascular endothelial cells from human eye's segments could be successful cultured,On the7-9th day ,the cells displayed monolayer growth and looked like"cobblestones", Immunohistochemically these cells were stained positively by â…§-Rag monoclonal antibody.The vascular endothelial cells had some degress of proliferation for experimental and control group in 3 days. The experimental group had no difference compared with the control group at 24 hours ( P>0.05).Group â…¢and group â…£had very significant difference compared with the control group ,groupâ… and group â…¡at 48 hours (P <0.01;group â…¢had very significant different compared with the control group and Other experimental groups. Conclusions :Certain concentration of bFGF has a very strong proliferative action on the vascular endothelial cells .In centation range of dose,bFGF significantly induced the endothelial proliferation in tro in a dose dependent manner,But exceed centain dose (80ng/ml ),bFGF significantly induced the endothelial proliferation can not increase.we guess,high dose bFGF maybe have some poisonous to the endothelial besides inducing proliferation. In all , bFGF significantly induced the endothelial proliferation, the concentration of 80ng/ml is themost strong . Neoangiogenesis induced in early by bFGF may open up ,new possibilities for the treatment of hydroxyapatite orbit implants .
