MTT and PCNA were used to detect the effects of resveratrolon proliferation of hepatocarcinoma and nomal cell line. The effects ofresveratrol on apoptosis of the four cell lines flow was detected bycytometry and TUNEL. Adhesive test, moving test and invasive test wereused to observe the effects of resveratrol on invasion ability ofhepatocarcinoma cell lines. And to tell the difference of the effects onthe three hepatocarcinoma cell lines. Results 12.5μmol/L resveratrol inhibited proliferation of HepG2(P﹤0.01), 25μmol/L resveratrol inhibited proliferation of Bel-7404 andSMMC-7721(P ﹤0.01), Once the concentration of resveratrol is upon100μmol/L, proliferation of normal cell L02 was inhibited byresveratrol(P﹤0.01). 25μmol/L resveratrol induced apoptosis of HepG2(P﹤0.01), and 50μmol/L Resveratrol induced apoptosis of Bel-7404(P﹤0.01), but the effect on apoptosis of SMMC-7721 was uncertain. As well, when the concentration is upon 100μmol/L, resveratrol could induceapoptosis of normal cell(P﹤0.01). 25μmol/L and 50μmol/L resveratrolcould inhibit the adhesion of Bel-7404(P﹤0.05; P﹤0.01) and SMMC-7721(P﹤0.01), and could inhibit the invasion of Bel-7404(P ﹤0.01) andSMMC-7721(P﹤0.01). However, the moving of the two cell lines was notinhibited. In the three tests, HepG2 was not inhibited by resveratrol. Conclusion Resveratrol inhibit the proliferation and induce the apoptosisof hepatocarcinoma cell lines, and inhibit the invasion ability ofhepatocarcinoma cell line through inhibiting adhesion to FN anddegradation of membrane matrix. The inhibition degree is different in thethree cell lines, and there is some cell selectivity in the effects onapoptosis of different hepatocarcinoma cell lines, so were the effect oninvasion. Resveratrol with high concentration(≥100μmol/L) inhibitproliferation of normal cell and induce apoptosis of normal cell of it. | | Keywords/Search Tags: | Resveratrol, Hepatocarcinoma cell lines, Proliferation, Apoptosis, Invasion | | Related items |
