Object: To investigate the effect of lysophosphatidic acid(LPA) andtransforming growth factorβ1 (TGFβ1 ) on the cell ultrastructure, proliferationand phagocytosis of rhesus retinal pigment epithelialium cells(rRPE) in vitro.Methods: Adult rRPE cells were harvested by enzyme dispase. The primaryrRPE cells was cultured and passaged in vitro and observed by transmissionelectron microscopy. The proliferation of rRPE cultured with growth factor wasobserved by MTT. The cell cycle of rRPE was observed by flow cytometry. And theexogenous pigment in cultured rRPE was detected by enzyme linkedimmunoassay.Result: The absorbance of the TGFβ1 group was lower than that of the controlgroup from the second day (P <0.05), but the LPA group was higher(P <0.05)than that. The G1 phrase of cell cycle of TGFβ1 group increased(P <0.05), butthe G2/M phrase decreased(P <0.05). And the G1 phrase of LPA group decreased(P<0.01) as the G2/M phrase increased(P <0.01). The phagocytosis of rRPE inthree groups had not statistically significant. (P >0.05). The ultrastructurechange of cell injury was not observed by transmission electronmicroscopy in the LPA group. The adverse change induced by TGFβ1 wasobserved, but there was no apoptosis characteristics.Conclussion: LPA promotes the cell proliferation of rRPE obviously. LPApromotes DNA synthesize in S phrase and cell devision in G2/M phrase. But TGFβ1showed the opposite effects. There were not adverse effects in LPA group,but some toxic action may be induced by TGFβ1. The mechanism need to bestudyed futher.
