OBJECTIVE: To explore the effects of levodopa on the therapy of rats in Parkinson' s disease(PD) in different drug delivery , and it' s neurotoxicity in the same dosage.METHODs: Make the rat model of PD and the successful rat models were assigned randomly into three groups (n=14) :A1, B1, C1. Also, the controlled groups named A2, B2, C2 were set up(n=14) at the same time. All rats in A1, B1, Clgroup were all treated with levodopa individually through gastrointestinal tract, subcutaneous part and brain ventricle. The rotation behaviour was induced by apomorphine at the same time in the next day after different levodopa delivery duration(2d, 4d, 6d, 8d) , and the number of rotation in 30 minutes was counted . At the last time when the rats were induced , initiating time, the maximal velocity and rotational duration were also counted. All the data were made statistics. After continuous drug delivery for 15 days , all rats were made internal fixation , then TH and HE staining were made. The positive cell stained by TH in nigra were counted and made statistics.RESULT:As compared with the controlled group, the rotation isreduced in different durations(p<0.05). But the data sh<^- no difference between the controlled group(p>0. 05). Initiating time is reduced and rotational duration is shorten (p<0. 05). The number of cell in right side is smaller contrasting with left sicU\ but which in the same side is no significance.CONCLUSION: The three ways of levodopa delivery are effect ive on treating PD rats. In the same therapy effect, the dosa^o of levodopa delivery through subcutance and ventricle are smaller, and the latter is smallest. In this studies, the neurotoxicity isn' t found and there' s significance in clinic practice of the last two ways of levodopa delivery.
