ObjectiveThis study was performed to investigate the effect about rhEPO on cell apoptosis, inflammatory response and brain edema around the traumatic brain zone and explore its protect role in traumatic brain injury. Methods1. Animal model 90 health male SD rats were randomly divided into three groups. Those were respectively sham groups, control groups and rhEPO groups. The model was made by improved Freeney's free weight traumatic brain injury. All animals were respectively rejected with saline or EPO 5000u/kg after the model was made. The animals of each group were killed after 12,48,120 hours of post-trauma. Those were respectively three 12h groups , three 48h groups and three 120h groups according to the killed time of post-trauma.2. Physiological parameter menstruation Contractive pressure, WBC, RBC and HCT were detected in all animals before they were killed.3. TUNEL and immunhistochemical detection Five SD rats of each group were killed and their brains were excluded after 12,48,120 hours of post-trauma. HE and TUNEL and immunhistochemical of MCP-1 and CD68 respectively stained four contiguous blades obtained at the core of traumatic brain zone after the samples were fixed. Then the number of the apoptic cells , MCP-1~+ and CD68~+ cells were detected.4. Detection of brain water content The other 5 SD rats of each group were killed and their brains were excluded after 12,48,120 hours of post-trauma. Then the brain water content of the injured hemisphere was detected.Results1. Multi-parameter of physiological conditions was no difference between EPO groupand control group (p>0.05).2. At each time, the number of the apoptic cells of rhEPO group was less than that of the control group around the traumatic brain injury lesion (p<0.01).3. At each time, the number of MCP-1+ and CD68+ cells of rhEPO group was less than that of the control group around the traumatic brain injury lesion (p<0.01).4. At each time, the brain water content of the injured hemisphere of rhEPO group was less than that of the control group (p<0.01).Conclusions1. There was no significant effect about rhEPO on multi-parameter of physiological conditions in a short period2. RhEPO can significantly decrease cell apoptosis around the traumatic brain injury lesion.3.RhEPO can significantly decrease inflammatory response around the traumatic brain injury lesion.4. RhEPO can significantly mitigate brain edema in traumatic brain injury...
