Objective To develop the new anti-leukemia medicine from the natural plants and to supply the theory for its application in clinic, the mechanism of anti-lymphocyte leukemia by Chinese traditional medicine, Euphorbia Ebracteolate Hayata,was sdudied.Methods The first step of the present study on the anti-leukemia mechanism of Euphorbia ebracteolate Hayata is to find the purity Euphorbia Ebracteolate Hayata and then to identify its breed. Then its availability components were distilled by water and 95% alcohol. The second step is to do a series of experiments about the anti-leukemia mechanism of the water-distilled components of Euphorbia Ebracteolate Hayata.Firstly, some toxicological experiments were done to compare the toxicity of the water-distilled components of Euphorbia ebracteolate Hayata and CP: The content of MDA in the liver and kidney of leukemia P388 bearing mice was mensurated. The peripheral blood micronucleus experiment, the sperm aberration experiment, the MTD experiment and the pathomorphological observation of the significant viscera of the mice were performed.Secondly, the lymphocytic leukemia P388 bearing mice acted as animal model, the research was carried out on the effect of the water-distilled components of Euphorbia Ebracteolate Hayata on inhibiting the malignantproliferation of WBC in peripheral blood and lymphocytic leukemia P388 cells in ascites, prolonging the survival time and inhibiting pathological changes of leukemia mice, enhancing the activity of anti-lipid peroxidation system,increasing the immunity of the body, inducing the apoptosis of leukemic cells.Results The results showed that the water-distilled components of Euphorbia Ebracteolate Hayata could significantly inhibits the malignant leucocyte proliferation induced by the attack of leukemic cells, prolongs the survival time of the lymphocytic leukemia P388 bearing mice, relives the spleen swell caused by leukemic cells and recovers the activity of SOD and GSH-PX in liver, indicating that Euphorbia Ebracteolate Hayata could retard the development of leukemia and increase the anti-lipid-peroxidation capability of the body.We have also found that the water-distilled components of Euphorbia Ebracteolate Hayata could inhibit the express of bcl-2 and promote the express of bax. In different dose groups, the influence degree on the expression of bcl-2/bax is also different. So, we assume that Euphorbia Ebracteolate Hayata be likely to inhibit apoptosis by influncing the expess of apoptosis gene, as bcl-2/bax.In the toxicology test of Euphorbia Ebracteolate Hayata, comparing with the controls the applied dosage has not found obvious toxicity to the body with the MTD>288g/kg. The significant sperm abnormity and mutation have not observed in mutation and sperm abnormity tests. Comparing with cyclophosphamide even the dosage increased to 77g/kg, each toxicity index is far lower than that of cyclophosphamide in routine dosage, showing that Euphorbia Ebracteolate Hayata is a secure and lower toxic natural medicine. However, there are some pathological changes in the 288g/kg dose group and no changes in other groups.Conclusion In conclusion, the anti-leukemia effect of Euphorbia Ebracteolate Hayata is reliable and the secure range is wide. The mechanism may be through inducing the tumor cell apoptosis but only killing them. Euphorbia Ebracteolate Hayata dispersed widely over our country with a low price. According merchandise investigation, 73% "lang du" merchandises is Euphorbia Ebracteolate Hayata and Euphorbia Fisheriana Stead. Euphorbia Ebracteolate Hayata is the main supply as "lang du", revealing the essentiality of the present research and the magnificent exploiting future of Euphorbia Ebracteolate Hayata in clinical application.
