Objective: To explore the the expression of PTTG and c-myc gene in acute leukemia (AL) and its relationship with acute leukemia. To explore the correlation between overexpression of PTTG and abnormal expression of oncogene c-myc in AL, and chemotherapy effects and prognostic factors of AL.Methods: Expression of PTTG and c-myc gene mRNA were detected in BMMNC from 46 patients with untreated acute leukemia , 15 patients who reached CR , 14 patients who failed and 10 normal controls ,by reverse transcriptase-polymerase chain reation (RT-PCR) .Results:1. Expression of PTTG and c-myc gene in BMMNC were significantly higher in untreated AL patients than in normal controls( P<0.05) .2. The expression of PTTG and c-myc gene in AML and ALL showed no significance ( P>0.05). The expression of PTTG in ALL was higher than in AML but there showed no significance ( P>0.05). The expression of c-myc in AML was higher than in ALL but there showed no significance (P>0.05).3. Expression of PTTG and c-myc gene were lower in patients who reached CR than in patients who failed ( P< 0.05).4. Expression of PTTG and c-myc gene before chemotherapy were lower in patients who reached CR than in patients who failed ( P<0.05).5. Expression of PTTG and c-myc gene were higher in untreated patients withabnormal karyotype than in untreated patients without abnormal karyotype ( P <0.05). Conclusion:1. PTTG and c-myc mRNA were overexpressed in patients with AL.2. Overexpression of PTTG and c-myc mRNA were related to abnormalkaryotype.3. The effects of chemotherapy in the AL patients with overexpression of PTTGand c-myc gene were poor.4. PTTG may be a prognostic factor in acute leukemia patients.5. Overexpression of PTTG and c-myc mRNA in AL patients may be relatedto the progression and pathogenesis of acute leukemia.
