Objective: To investigate the inhibitory effects on invasion and metastasis of gastric cancer and possible clinic application of RAGE anti-sense oligoxydeonucleotide(AsODN) through observing the effects of RAGE AsODN on the motility of gastric carcinoma cell lines- MKN-28 cell. Methods: MKN-28 gastric carcinoma cells were cultured with RPMI-1640 media. Cultured MKN-28 cells were designed into 1.5μM RAGE AsODN group , 3. 0 μM RAGE AsODN group , 4. 5μM RAGE AsODN group , 6.0μM RAGE AsODN group , 3. 0u M sense oligoxydeonucleotide (SODN) group and the control group. The effects of3. 0μM RAGE AsODN on cell motility was evaluated by wound healing assay and millicell chamber assay, The effects of different RAGE AsODN on MKN-28 cells was measured by millicell chamber assay; the expression of RAGE protein in MKN-28 cells was detected by sp immunohistochemistry.Result: Treated with 3.0μM RAGE AsODN 12, 24, 48 hours, we found that there were almost little cell nest in the noncell area, however, in 12, 24, 48 hours, many scattered tumor cells can be observed in the noncell area of the control groups, compared with the 0 hour, the cells on the boarderline have become multiangular ones, and the expanding trend was obvious. many cell nests can also be observed in 12, 24, 48 hours in SODN group . RAGE AsODN could remarkly suppress the molitily of MKN-28 cell contrasted with the control group by millicell assay(P <0. 01), but there were not statistically significant between the SODN and the control groups (P>0.05).
