Objectives In this study, on the model of Middle Cerebral Artery Occlusion in Wistar rats, the neuronal ischemia situation was investigated by using hematoxylin and toluidine blue stain and the endogenous neural stem cells regeneration phenomena with different treatments were observed by using immunohistochemistry. Our purpose is to investigate the neurogenesis significance of endogenous neural stem cells.Methods We used Cauterization method to build the MCAO model in Wistar rats, and then divided into 6 groups, including normal, sham, Jingxue blood bleeding, Chinese herb, Cyclosporine A and control group, 5 rats in each group. Frozen sections from the brain tissues were preparied in immunohistochemistry studies to investigate IHC result, such as Nestin, BrdU, PCNA, GFAP and NSE and also in the hematoxylin and toluidine blue stain to observe the pathological changes.Results In the Jingxue blood bleeding treatment group, the immunohistochemistry positive cells counting are more than that of control group apparently whichever the BrdU, Nestin and NSE. In the Chinese herb treatment group, the immunohistochemistry positive cells counting are more than that of control group apparently whichever the BrdU, Nestin and NSE. In the Cyclosporine A treatment group, the immunohistochemistry positive cells counting has no significant difference compared with control group.Conclusion1. The blood bleeding therapeutics can offer the neural repairing and prevention effects after Middle Cerebral Artery Occlusion.2. The given Chinese herb therapeutics can promote the proliferation of neural stem cells after Middle Cerebral Artery Occlusion.3. Cell mediated immunity has no dependability with the proliferation spark of neural stem cells after Middle Cerebral Artery Occlusion.
