The Study Of IFN-γ And IL-4 Secreted From T Helper Cell In Endometriosis

Objective: In, this study, we examine the concentrations of interferon- γ (IFN-γ ) and interleukin-4 (IL-4) secreted by T helper cell(Th cell) in the peritoneal fluid and peripheral blood of endometriosis patients. We also calculate the ratio of IFN—γ / IL-4, and explore their role in the endometriosis.Methods: 24 women with histopathological diagnosed endometriosis after operation at stage Ⅰ-Ⅳ according to the criteria by Revised American Fertility Society were selected as endometriosis group, 24 women with leiomyoma or benign ovarian tumor were selected as control group. Isolating and culturing Th cell in the peritoneal fluid and peripheral blood, the concentrations of IFN-γ and IL-4 in culture media were determined by enzyme-linked immunosorbent assay(ELISA).Result: The mean IFN- y concentration secreted by Th cell in peripheral blood of endometriosis group is 337.63 ±62. 94ng/L(X ± S , others are same),significantly lower than 440.83 ± 67.67ng/L of control group(P<0.01); The concentration in peritoneal fluid is 62.47 ± 22. 12ng/Lsignificantly lower than 103.83 ± 30.84ng/L of control group (P<0. 01) ; The mean IFN- Y concentration in both peripheral blood and peritoneal fluid is no difference in menstrual cycle and disease' s state(P>0.05). The mean IL-4 concentration secreted by Th cell in peripheral blood of endometriosis group is 9. 04 + 3. 19ng/L significantly higher than 7.11 ± 2. 39ng/L of control group(P<0. 05) ; The concentration in peritoneal fluid is 1.73 ± 0. 73ng/L significantly higher than 1.225 ± 0.43ng/L of control group(P<0.05) ; The concentration in peritoneal fluid of proliferative phase is lower than secretory phase(P<0.05) ; there is no difference of disease' s state in both peripheral blood and peritoneal fluid. The ratio of IFN-V / IL-4 in peripheral blood of endometriosis group is 39. 51 ±7.34 significantly lower than 65. 86 ±12. 77 of control group(P<0. 01) ; The ratio in peritoneal fluid is 37.93 + 8.30 significantly lower than 86.68 ± 12.77 of control group(P<0. 01).Conclusion: The Th cell of endometriosis group secreting IFN- y is declined but secreting IL-4 is significantly strengthened.
