Objective: There is long history in mammary cancer treated by traditional Chinese medicine. As a kind of rare Chinese medicinal herbs, CS can support body constitution. Clinical and experimental study indicated that CS also has good anti-tumor effect. In order to understand the anti-tumor effect of CS and examine the induction of apoptosis in cancer cells by CS, we cultured MCF-7 breast cells, use molecular biology and immuno-histochemistry to investigate whether CS can induce apoptosis of breast cancer cell line and elucidate the regulation of CS on Bcl-2 family genes.Method:1. Cultured MCF-7 cells were treated with different concentrations of CS for different times: 0.1ug/ml, 1.0ug/ml, 10ug/ml, 20ug/ml, 50ug/ml, 100ug/ml for 24, 48, 72, 96, 120 h, MTT assay was used to measured the anti-proliferative effect of CS;2. MCF-7 cells were treated with lOug/mK 20ug/ml> 5Oug/ml of CS for 48 h, cell cycle and apoptosis of MCF-7 cells were analyzed by flow cytometry (FCM); MCF-7 cells were treated with 20ug/mh 50ug/ml of CS for 48 h, apoptosis induction was also proved by agarose gel electrophoresis;3. MCF-7 cells were treated with lOug/mK 50ug/ml ofCS for 48 h, alteration of protein of Bcl-2 and Bax genes was detected by immunohistochemical techniques.Results:1. MTT assay showed CS could inhibit the growth of MCF-7 cells in a dose/time dependent manner, the anti-proliferative effect was more obvious from l.Oug/ml to 50ug/ml during 24~96h;2.
