Hepatocellular carcinoma (HCC) is one of the ten most common cancers in humans with over 500,000-1,000,000 new cases diagnosed worldwide each year. A number of etiological factors, particularly hepatitis B virus (HBV) infection, are involved in the occurrence and progression of HCC. Epidemiologically, 80% of all HCC cases have been associated with long-standing HBV infection. Chronic infection with HBV is associated with an increased (100-200 times) risk of developing liver cancer. HBV commonly infects neither cultured cells nor ordinary laboratory animals (only chimpanzee being a kind of sensitive experimental animal) . This has limited the research on HBV pathogenesis to some extent. HBV transgenic mice have provided insight into the mechanism of HBV pathogenesis and hepatocarcinogenesis.The biochemical parameters were assistance to estimate the trauma of liver. The biochemical parameters of the HBV transgenic mice were detected respectively at the age of 2,6,12,18,24 months,the activities of ALT, CHO, TG, TP were significantly different compared with wide type control mice, which indicated the liver of the HBV transgenic mice had already damnified in early time.Distinct pathological changes appeared with age increasing, lymphocyte infiltration, fatty degeneration and atypical hyperplasia were observed inthe liver of transgenic mice at 6 and 12 months. HCCs happened at 18 and 24 months, and metastasis of HCC was found in elderly transgenic mice.We use the method of OPS to preserve embryos in vitrification solution, we found that the average numbers of embryo production of P21+/+, P21HBsAg/HBsAgand p21HBx/HBx were 27.89,15.56 and 9.14 respectively, the recovery rates were 88.7%, 81.1% and 80.3% respectively, the development rates were 82.6%, 79.4% and 56.6% respectively after thawing. The results indicated that the embryos number were significantly lower than wild type mice, the recovery rates have no significantly difference among the three genotype mice, the development rates of p21HBx/HBx were significantly lower than the ones of P21+/+, p21HBsAg/HBsAg We gain 5 transgenic mice through embryos transplant. The results indicated that the method of vitrfication cryopreservation embryos of transgenic mice is successful.
