Background: Approximately 120 million people all over the world have diabetes. Among persons diagnosed as having diabetes mellitus, approximately 3-4% of individuals currently have foot ulcers or deep infections. Fifteen percent develop foot ulcers during their lifetimes. Foot ulcers and their complications are an important cause of morbidity and mortality in diabetes. Diabetic foot ulcers (DFUs) precede 85% of nontraumatic lower-extremity amputations (LEAs). At 2 years following transtibial amputation, 36% have died. Diabetic foot ulcers have been neglected in health-care research and planning , which lead to an absence of suitable animal model of diabetic ulcers and hinder the research of diabetic ulcers.Objective: To prepare rat model of diabetic ulcers and investigate its biological characteristics.Methods: Recent research demonstrated that Ischemia-reperfusion injury play a role in chronic wounds induced by diabetic. According to the principle of Ischemia-reperfusion injury, we have prepared diabetic ulcers in diabetic rat by the method of Cyclical Magnetic Compression in this study. Protocol is following: firstly, plant magnetic disks under skin of rat, then induce diabetes Mellitus by injected with 50mg/kg STZ after wound healing. After formation of DM, diabetic ulcer was induced by the method of Cyclical" Magnetic Compression (Magnet Application Protocol is following: ischemic times is 2 hours, reperfusion times is 30 minutes, three cycles per day).
