| The gastric carcinoma is a common digestive syserm malignant tumor,the morbidity of which is high,at the same time its treatment effect is not so good.It is one of the common malignant tumor that serierously affect people's health.But it is not clear for its pathogenesy up to now.Recently much research sign that it is related between COX-2 and tomor.It offers the new points for gastric cancer. Cyclooxygenases (COXes), which are the key enzymes in prostaglandin (PG) biosynthesis,.cyclooxygenases have at least 2 isoenzymes in the mammal, namely COX-1 and COX-2. COX-1 is housekeeper gene,localizating 9q32-33.3 chromome, about 22.5kb,coding 599-600 amino-acid residue,including 23 or 24 signal peptides that it is consist of amion-acid residue,composing 11 extrons and 10 introns.The COX-1 is rating-limited enzyme that resting-cell synthesis PG. It can maintain normal physiologic function,for protecting gastric mucosa,regulating renal blood flow,controlling platelet aggregation et al. The COX-1 express in most normal tissue.The COX-2 is guiding-type enzyme,locating1q25.2-25.3 chromome,, composing 10 extrons and 9 introns,about 8.3kb,coding 604 polypeptides amino-acid residue,including 23 or 24 signal peptides that it is consist of amion-acid residue, including 17 signal peptides that it is consist of amion-acid residue,The COX-2 does not expess in normal tissue,but up-regulation when induced growth factor and cytokines. There are three methods to detect COX-2:1,detecting COX-2mRNA,as if Northern blots, Reverse-Transcription Polymerase Chain Reactin(RT-PCR);2,detecting COX-2 protein,as if immuno-histochemistry SP,western-blotting; 3,Detecting COX-2 products or correlated enzyme to dertermin expression of COX-2,for PGE2,MMP-2,MT-MMP-1.In this study, we used immunohistochemistry method to detect the expression of COX-2 in 40 human gastric cancer and 12 nomal gastric mucosa ,and to understand the relation between the expression of COX-2 and the pathological parameter. The experiment chooses 40 cases gastric cancer treated in china-japan Union Hospital of Ji Lin University..Among them 18 cases are males, 22 cases are females,the age between 37～78 year old,the average age is 58.5.All cases depends on pathology diagnosis. histologyical well and moderately differentiated adenoma 9 cases, poorly differentiated adenoma 31 cases, lymph-node metastasis 28 cases, non-lymph-node metastasis 12 cases;TNM stage: I3 cases,Ⅱ8 cases,Ⅲ20 cases, Ⅳ9 cases.The patient do not treat by chemotherapy or radiotherapy or NSAIDs or hormone. The 12 cases normal gastric mucosa tissues are all from the same specimens as control,which is from carcinoma tissues about 6-8cm(confirmed by pathohistology). The result of the immunohistochemistry indicates as following: COX-2 positive 35 cases,low expression 10 cases,high expression 25 cases,with the total positive rate 80%,and of the control shows us that:negative 9 cases,low expression 3 cases,high expression 0 case,with the total positive 25%.
