Background and objective: Now, most of chemotherapy medicines have owned basic and clinic experiment data through many years study. Actinomycin is the newest chemotherapy medicines, so we are short of their interrelated experimental data of chemotherapy. This experiment will start to explore interrelated chemotherapy data of Actinomycin from both regular experimental animal degree and cell degree. Not only can the research supply indispensable experimental proof for framing chronochemotherapy in clinic, but also it is the first and the second parts of series studies.Methods: The experiment was divided into two parts. In part one, 140 4-week-age BALB/C rats of 20±2g weight were divided randomly into seven groups according to their sex. Every group had 20 rats in halves of male and female. They were fed with detached cages according to sex. Six groups were randomly appointed to 0 hour group, 4 hour group, 8 hour group, 12 hour group, 16 hour group, 20 hour group according to the six time of injection of 0: 00, 04: 00, 08: 00, 12: 00, 16: 00, 20: 00. The remainder group was left for blank controlled group. They were synchronized with natural and artificial lighting regimen with 07: 00-19: 00 hours in light and 19: 00-07: 00 hours in dark for at least 3 weeks. Afterwards, these experimental groups' rats were injected 235.4ug/kg Actinomycin in peritoneal cavity strictly according to the designed time, while those controlled groups rats were injected 0.24ml physiological brine. After 10 days, we drowned their blood sample through snipping the tails and mensurated their number of white blood cell. In part two, experimental process was basically the same to the first part except for injecting 235.4ug/kg Actinomycin in peritoneal cavity. Afterwards, the number of died rats was recorded in 72 hours from injecting medicine.Results: (1). There was significant difference about the number of white blood cell in all groups rats' blood through SPSS test (P<0.001). there was significant circadian rhythm about the number change of white blood cell in all groups rats' blood through Cosinor software analysis (P<0.001).
