[Abstract] Objective To study the feasibility of the transplantation with mixed hematopoietic stem cells of the metaphase embryo SD rats and to search for the new source of hematopoietic stem cells. Methods The hematopoietic stem cells (HSC,8×10~7 MNCs) from one embryo SD rat (gestational age 14~17d) were transplanted into every one of 40 female SD rats of experiment group and the HSC (8×10~7 MNCs) from one grown male SD rats'marrow were transplanted into every one of 10 female SD rats of BMT group by the tail vein after these rats were sublethally irradiated with 5GY (60)~Co . At the same time, 10 female SD rats of irradiated group were sublethally irradiated with 5GY (60)~Co . In 21 days after hematopoietic stem cells transplantations , the following items were observed : common condition ,peripheral blood, MNCs and CFU-GM from the recipient rate'bone marrow cells ,GVHD and the Transplanted evidence (SRY gene). Results The number of MNCs (×10~7) from an embryo rat (gestational age 15~17d , 11.5±2.8 ,13.4±2.6 and 13.7±2.7 ) is the same as that from a grown rats'marrow ( 18.6±2.7, P>0.05),it is enough to be transplantated to one grown rat .The number (/1×10~5MNCs)of CFU-GM of the HSC from an embryo rat (gestational age 13~17d ,180.6±25.1 ,184.3±31.1 ,188.8±28.8,178.3±26.0 ,179.8±32.0) is significantly higher than that from a grown rats'marrow ( 62.7±13.3, P<0.05),these results show that the multiplicating ability of HSC from an embryo rat(gestational age 13~17d) is stronger than that from a grown rats'marrow cells . 26 cases survived in 40 rats in 21 days after the embryo rat(gestational age 14~17d) HSCT and 21 rats'hemopoietic system.were repopulated smoothly. 28 cases occurred GVHD(Ⅰo~Ⅱo ) in 40 rats. 7 cases survived in 10 rats in 21 days after the embryo rat(gestational age 15d) HSCT , the transplanted evidence (SRY gene) was detected in 6 cases's marrow cells. Conclusion The number (×10~7)of HSC from an embryo rat (gestational age 15~17d) is the same as that from a grown rats'marrow and enough to be transplantated to a grown rat . The multiplicating ability of HSC from an embryo rat (gestational age 13~17d) is stronger than that from the grown rats'marrow . The HSC from an embryo rat can be engrafted a grown rat's marrow which suffered from sublethal irradiation with 5GY 60Co and repopulate the hemopoietic system of this rat.. Embryo hematopoietic stem cells possibly provide a new and promising source of hematopoietic stem cells.
