| Objective: To investigate the expression of cytolkines:basic fibroblastgrowth factor(bFGF),transforming growth factor-beta-1(TGF-β1)and theproapoptotic gene bax and antiapoptotic gene bcl-2 in the amniochorionwith premature rupture of membranes ( PROM ) and to probe thepathogenesis of PROM. Methods: Fresh amniochrionic membranes were collected randomlyfrom the following groups of women (gestation age ≥ 28 weeks) :(1)30women after vaginal delivery with PROM as the study group, includingpreterm labor 16 cases and term labor 14 cases,(2)30 women after vaginaldelivery with intact membranes as the control group,including pretermlabor 11 cases and term labor 19 cases. All membranes were processed bywashing,fixing and immunohistochemistrical staining. Observing thelocation in the amniochorion through light microscope and caculatingpositive average integral calculus oculus dehtor. Result: 1.Proapoptotic protein bax are located in the plasma of all celltypes:trophoblast ,epithelial,mesenchymal and fibroblast ,but bcl-2 only can beobserved in chorionic syncytiotrophoblast. bFGF and TGF- β 1 arelocateded in the plasma of all celltypes. 2.The positive average integral calculus oculus dehtor of bax,bFGFand TGF-β1 in study group were respectively 0.086±0.021 ,0.051±0.012,0.059±0.013,in control group were respectively 0.046±0.018,0.098±0.027, 0.119±0.032, the expression of bax in study group wassignificantly higher than that in control group(P<0.001) while theexprssion of bFGF and TGF-β1 was significantly lower than those ofcontrol group (P<0.001,0.001,respectively); the expression of bcl-2 insyncytiotrophoblst was not significantly differential in both groups(0.058±0.020,0.050±0.014 ,P>0.05).Comparing with preterm and term labor inboth groups respectively,the expression of all marks were not significantlydifferential(P>0.05). 3. Bax in study group has negative correlation with bFGF andTGF-β1(r=-0.616,-0.489,P<0.05, 0.05, respectively),and bax in controlgroup does not have relationship to bFGF and TGF-β1 (r=-0.302, -0.137,P>0.05,0.05, respectively); bFGF does not have relationship to TGF-β1 inboth groups (r=0.359,0.158,P>0.05,0.05,respectively). Conclusion: Bax are located in the all celltypes in amniochorionwhile bcl-2 only in syncytiotrophoblst, bax and bcl-2 are the mainapoptosis regulation gene in fetal membranes. the occurrence of PROMclosely relate to the apotosis; TGF-β1 and bFGF can be excreted byamniochorial cells,and the expression of them have relation to proapoptoticgene bax,may have important function in the occurrence of PROM. |
