Objectives: To approach the effects and mechanisms on stimulatingthe regeneration of corneal allografts' nerves by using PEDF eye dropsafter penetrating keratoplasty on rats. Methods: Penetrating keratoplasty (PKP) was performedorthotopically from Wistar rats to SD rat' recipients. 72 SD rats wererandomly assigned to two groups-experimental group and control groupafter PKP. Experimental group was treated with PEDF eye drops, Controlgroup was treated with escipient eye drops. Both experimental group andcontrol group were divided into 6 groups-one week, two weeks, threeweeks, one month, two months, three months. There were 6 animals ineach group. Use Cochet & Bonnet aesthesiometer to detect the recovery ofthe allografts' nerves. Detect the regeneration of the allografts' nerves by invivo confocal microscope. 6 animals in both groups were killedrespectively at the 1st week, 2nd weeks, 3rd weeks, 1st month, 2nd monthsand 3rd months postoperatively, staining with hemotoxylin and eosin (HE).PEDF expression was examined by immunohistochemistry. Results: 1. The recover time of the allografts' nerves in experimentalgroup was obviously shorter than the control group. 2. Contrast with thecontrol group, the amount of the allografts' nerves was obviously increasedin experimental group; the growing speed of the allografts' nerves inexperimental group was obviously more quickly than the control group. 3.Significantly increased PEDF expression was also observed inexperimental group as compared with control group byimmunohistochemistory staining. 4. In comparing with the control group,the repair of the corneal epithelium in the experimental group wasincreased significantly. Conclusions:1. Topical application of PEDF eye drops can stimulatethe regeneration of the corneal nerves of allografts. 2. Topical applicationof PEDF eye drops can accelerate the repair of the corneal feeling. 3. PEDFcan enhance PEDF expression in corneal allografts.
