Qilingyiganjian (QLYGJ) is a mixture of some herb, such as Huangqi, Lingzhi, Ezhu, Banzhilian, etc. It has functions of cleaning away heat and toxic material, restoring normal functioning of the body to consolidate the constitution and resolving mass. It has been proved that QLYGJ has strong anti-cancer effect on primary hepatic carcinoma during our two years'study [1][2]. The technique of CO2 Supercritical Fluid Extraction (CO2-SFE), which is infant and progressive nowadays, attracts more and more respect, especially in crude plant extraction, and its perspective is fairly broad. In contrast with traditional extraction, that is, gas-distillation and organic solvent extraction, The technique of CO2-SFE has so many advantages that it is widely used in extracting volatile oil, alkaloid, coumarin, lignin, flavone compound, sugar and glucoside, etc. Above all, it can realize the separation of the valid element and poisonous material. Telomerase is a ribonucleoprotein complex, which has the character of reverse-transcriptase. It can synthesize and prolong telomere repeat sequence, that is, TTAGGG, by its own template. By this means, cells that have positive telomerase can avoid replicate senescence, and get to immortalization. Over 85% hepatic carcinoma exists this mechanism, human telomerase comprises two subunits: human telomerase RNA component (hTR) and human telomerase reverse transcriptase (hTERT), the former provides an RNA template and expresses in most cells. So, it has no differentiation. However, the latter only express in telomerase positive cell. Therefore, studies on TERT have been one of the most important topics in carcinoma research. Some studies show that some herb such as Ezhu and Lingzhi can restrict the growth of cancer by downregulating TERT. Through serologic pharmacological test, using cyclophamide (Cy) as positive control, the morphologic changes of H22 cells treated with pharmic serum were observed under phase-contrast microscope, then, the resist tumor rate by MTT was detected. Lastly, the expression of TERT in H22 treated by pharmic serum was studied, and the findings show as follows: 1. The morphologic observations on H22 treated by pharmic serum under phase-contrast microscope show that: The cells treated by negative serum grew well, they were round and reflected bright light, and the nucleus were big and the intramembrane was clear. While the cells treated with pharmic serum were dying. The were crimpy and dark, and they didn't reflect bright light. Even many fragments appeared. 2. Detection of tumor resist rate by MTT: The OD numerical value of these five group is as follows: That of the group of liquid of CO2-SFE was 0.3094±0.016, that of the group of high dose QLYGJ was 0.3514±0.020, that of the group of the low dose of QLYGJ was 0.4244±0.030, that of the group of Cy was 0.3880±0.0240, while that of the group of blank serum was 0.4986±0.031. The result shows that: by method of One-Way ANOVA, All of the OD numerical value of these five group are not all the same (P<0.01). By method of Dunnett-t test, the difference of the four groups of cells treated withpharmic serum has statistic sense (P<0.01) in contrast with negative serum. By method of LSD test, the difference of either two of these five groups makes statistic sense (big dose and Cy, Cy and low dose, P<0.05, and the others P<0.01). That is, the effect of suppress tumor of the five group are different, the groups whose suppress tumor rate from strong to ebb are as follows: the group of liquid of CO2-SFE, the group of big dose, the group of Cy, the group of low dose, and their suppress tumor rates are as follows: 37.9%, 29.5%, 22.1%, 14.8%. 3. The expression of TERT through immunohistochemical staining: All of the five groups had positive expression. By K Independent samples' rank test (method of Kruskal-Wallis), the result shows that: The levels of the expression of TERT on H22 cells treated with five group of serum are not all the same (P<0.01).
