| Objective to study on the effects of radiofrequency energy and treatments time on chondrocytes and matrix of fibrillated articular cartilage.Methods Set up 3 patterns of the bovine knee's mechanically traumatized cartilage, choose 2 experimental regions in each bovine's knee, cut each experimental region into 4 parts sized 1cm×1cm. then choose 3 parts as experimental groups treated respectively for 1 min by 15W,40W,70W, take the untreated part as control group, so there are 6 experimental regions, 4 groups in each region, 24 samples in all, harvest full-thickness cartilage from each part as explants, divide each explant into three parts and treat them respectively by HE stain ,flurescein diacetate and propidium iodide stain, test the release of glycosaminoglycans to assess the relation between the radiofrequency energy and the damage of Articular Cartilage. meantime, use the same methods and designs to set up another 3 experimental degenerated articular cartilage patterns with different parameter, lOsecs, 30secs and 60secs respectively by 40W radiofrequency energy and the grouping, the method of treatment is also the same to assess the relation between the time of treatment and the damages of Articular Cartilage.Results of HE stain: with the increase of radiofrequency energy, the death of Chondrocytes increases significantly, to group 15W, no apparent difference from control group (p>0.05), while to group 40W and 70W distinguished difference (p<0.05). Meanwhile, with the increase of time, the death of Chondrocytes increases significantly, to 10 secs group, no difference from control group (p>0.05) while to 30secs and 60 secs groups, distinguished difference from control group(p<0.05). results of flurescein diacetate and propidium iodide stain, with the increase of radiofrequency energy, the death of Chondrocytes increases significantly, to 15W group , no apparent difference from control group (p>0.05), to 40W group and 70W group, distinguished difference from control group (p<0.05), meanwhile with the increase of time, the death of Chondrocytes increases significantly, to10secs group, 30secs group and 60 secs group, distinguished difference from control group (p<0.05). Results of the release of glycosaminoglycans, with the increase of radiofrequency energy, the release of glycosaminoglycans of the first two days decreases significantly, to 15W group no difference from control group (p>0.05) while to 40W group and 70W group, distinguished difference from control group (p<0.05), on the third day, though no obvious difference in GAG release among the mentioned groups, compared with control groups, GAG release decreases in the experimental groups.
