Chondrotin Sulfate's Influence To The Proliferation And The Differentiation Of HL60 Cell And It's Pre-applied Study

purpose Observing the influence of ChS to the proliferation and the differentiation of HL60 cell, inquiring the ChS's application value in chemotherapy of malignant tumor.Method HL60 cells with same density was divided into 5 groups at random , added with 0 μ g/ml, 5 μg/ml, 25 μg/ml, 50 μg/ml and 75 μ g/ml ChS. (1) Hatching for 24 hours, proceed cell count and compare the different clone-growth rate with each other groups. (2)Add ADR to each groups after hatching for 24 hours, then hatching for 12 hours sequentially, compare the different surviving rate of cell with each other groups. (3)Hatching for 5 days, measure the cell's NBT deoxidization ability, compare the difference with each other groups. (4)Hatching for 5 days, add ink to each of the groups, observe and compare the different gobbling ability of cell with each other groups by high-scale microscope. (5)Hatching for 5 days, measure the acid phosphatase activity, compare the different with each other groups.Result 1 .All the concentration of ChS can increase the clone-growth rate of cell obviously (P<0.01), that is positively related to ChS's concentration in a limited range, but the density of cell shown a little difference when the concentration increased above 50μg/ml (P>0.05). 2.After ADR had been added, the cell's surviving rate of each ChS groups shown a decrease in different degree, especially the ChS concentration above 25 μg/ml (P<0.01). 3.NBT deoxidization ability and acid phosphatase activity of each ChS groups boosted up obviously (P<0.05), all the ChS groups displayed a strong gobbling ability.Conclusion 1.ChS can accelerate HL60 cell's proliferation; 2.ChS can increase the sensibility of HL60 cell to ADR and boost obviously up the kill effect of ADR to HL60 cell; 3.ChS can promote the differentiation of HL60 cell.
