Objective To study the expression and significance of aromatase protein and messenger ribonucleic acid , leptin messenger ribonucleic acid and leptin receptor messenger ribonucleic acid in endometrial carcinoma.Methods The expressions of aromatase , leptin and leptin receptor mRNA in 35 endometrial carcinoma tissues( including 23 cases with endometrial tissues near the carcinoma ) and 14 human normal endometrial tissues were detected by reverse transcription polymerase chain reaction (RT-PCR) .The expression of aromatase protein was detected by immunohistochemical streptavidin peroxidase technique . Results The expression of aromatase mRNA was 51.43 % in endometrial carcinoma and 78.26% in endometrial tissues near carcinoma. For aromatase protein the percentage was 45.71% and 69.57% respectively. Their differences were significant (P<0.05) and both were obviously higher than 0.00% in normal endometrium (P<0.01). There were no significant differences for the expressions of leptin and leptin receptor mRNA in endometrial carcinoma , endometrial tissues near carcinoma and normal endometrium (P>0.05) . The expression of leptin mRNA was different between endometrioid adenocarcinoma and the other histological types .The expressions of aromatase and leptin receptor mRNA did not show correlation with such factors as histological type , clinical staging and histological grading .Conclusion The distinguished expressions of aromatase mRNA and it's protein between endometrial carcinoma and normal endometrium suggest that aromatase plays an important role in the genesis of endometrial carcinoma .The high expression levels for leptin and its receptor in endometrial carcinoma and normal endometrium suggest that they may not involve in the genesis and development of endometrial carcinoma.
