PrefaceAlong with the increasing of the age of population in the world and the changing of life style, the cerebral vascular disease ( CVD) has become one of three diseases by causing death, deformity and affecting the level of life seriously. Currently, besides the medicine therapy, the research of the treatment of various physical factors becomes more and more. The effect of ultrashort wave on cerebral ischemia and reperfusion is reported less. Although it has many study in rotated magnetic fields therapy, the effect is uncertain . The therapeutic mechanism of the two methods is uncertain, too. Recently, it has reported many of the effect of the medicine on cerebral infarction. Physical therapy is always used in the period of recovery. We will investigate the effect and mechanism of ultra-short waves ( USW) treatment and rotated magnetic fields therapy ( RM) on the brain following transcient focal cerebral ischemia and reperfusion in rats. Rat model of focal cerebral ischemia and reperfusion were induced in the experiment , observing the effect of ultrashort wave and rotated magnetic fields therapy on hydrocephalus cohtent, cerebral infarct volume and histological expressional analysis of Bcl -2 and Bax.To investigate the effect and mechanism of ultrashort waves treatment and rotated magnetic fields therapy on the brain following transcient focal cerebral ischemia and reperfusion in rats.The Object and Method of Experiment1. The Object52 Wistar rats are, half in male and half in female, weighed 270 - 330g, 2. 5 -3 months old, made the model of focal cerebral ischemia and reperfusion by intraluminal filament occlusion. The entering standard is the rat model is graded 2 points by 5 levels grade made by Zea Longa, i. e. rats turn around to another side of focus when walking. 52 rats are randomly divided into sham operation group (n =9) ,ischemia reperfusion antrol group( n = 15) , ultrashort wave treatment group ( USW group for short, n = 14) and rotated magnetic field treatment group( RM group for short,n = 14).2. MethodUSW group: The electrotherapeutical instrument of ultrashort wave is the one used on five sense organs. The diameter of electrode is 4cm. The electrodes are contraposition , its surface is apart from the head of the rat about 2cm. The instrument is turned to apyretic, i. e. turning to the first gear, the power of output is 8.21W. Rats are treated about 10 min after cerebral ischemia and reperfusion 18h.RM group; The instrument of rotated magnetic fields therapy has a head (dia. 6cm) including two magnets (the position of N pole and S pole is apposition ) ,the disease of the center of the two magnets is 2.7 cm. The intensity of the magnetic field is 0.09T and the rotate speed is 2500r/min. The distance between the heads of instrument and rat is 0. 6cm. Rats are treated about 10 min after cerebral ischemia and reperfusion 18h.Antrol group: The method of therapy is the same to USW group and RM group, but the instrument of USW has no output and the one of RM has no magnet.3. The Index of Measuration (1) Hydrocephalus ContentThe rats are killed by decollation at 24 hours after ischemia and reperfusion. The brain is weighed on the scale ( definition, 1/10000) for wet weight, and for dry weight after dried in the oven (120 ). The hydrocephalus content comes from the Eliot formula, ( wet weight—dry weight)/ wet weight x 100%.(2)TTC DyeThe killing method is the same to the above. The brain is divided into many segments which is about 2mm in thickness. The segments of brain are put into 2% TTC solution for about 30min avoided the light in the 37°C flume, then into 10% formaldehyde solution for fixation. The infracting part are taken pictures and analysed by the system of image analysis for cerebral infarct volume.(3) Expression of Bcl - 2, BaxThe rats are perfused with 4% polyformaldehyde solution for fixation before dying. Take out the brain and take one segment(2mm in thickness) at where optic nerve is across. The segment is embedded in the olefin. Cut the segment into thin slice about 5 (xm thick. The thin slices are dyed by the immunohistolog-ical chemic method(the degree of pure,Bcl -2 and Bax are all 1:100). Under the microscope (400 multiple) , take 4 different parts in each thin slice and analyses the average gray value.(4) Statistical analysisThe results are given as the mean ± SD (x ± s). Groups are carried out by t test.Result1. Hydrocephalus ContentThe hydrocephalus content in diseased side brain of USW group (79. 390 1. 055) % is decreasing than antrol group ( 81. 393 ± 0. 825 ) % ( p < 0. 05) apparently, the one of RM group ( 80. 873 ± 1. 966) % is decreasing than antrol group, but the difference is not obvious( p >0. 05). The one of sham operation group (78. 528 ± 0. 271) % is decreasing than antrol group apparently ( p < 0. 01).2. Cerebral Infarct VolumeThe cerebral infarct volume of USW group (7. 97 ± 3. 21) % is decreasing than antrol group (14. 98 ± 5. 92) % apparently( P < 0. 05) , the one of RM group (10. 33 ±4. 73 ) % is decreasing than antrol group , but the difference is not apparently( P > 0.05). The sham operation group hasno infarction.3. Expression of Bcl - 2 and BaxThe average gray values of Bcl - 2 of USW group and RM group (136.112 ±2.654 ) (134.715 ± 2.028 ) are all higher than antrol group (131.434 ± 1. 578 ) ( p < 0. 01, p < 0. 01). The expression values of Bax of the two treatment groups (112. 974 ± 2. 225 ) (112. 862 ± 4. 170 ) are all smaller than antrol group (119.452 ±4.360 )(p<0.01, p<0.01), but the difference of expression of Bcl - 2, Bax between the two treatment groups is not apparently ( P > 0. 05 ).DiscussionIt confirms that the ultrashort wave (apyrexia) can decrease the hydroceph-alus content and cerebral infarct volume, when used on the treatment of injury of cerebral ischemia and reperfusion. It is helpful to recover the function of the ce-rebra. At the same time, USW can increase the expression of Bcl - 2 protein which can inhibit the out - break of apoptosis, and decrease the expression of Bax protein which can promote the occurrence of apoptosis. It confirmed that USW is effective on treating the injury of cerebral ischemia and reperfusion on the level of controlling apoptosis.RM treatment based on the condition of our trial can decrease the hydro-cephalus content and cerebral infarct volume,but the decrease is not apparently. Considering the reasons are the following. The first, the quantity of sample perhaps is much less. The second, the conditions of our trial are not compared with the ones of other trials. In our trial, the position of treatment is head, where the flow of the blood is weak, the time of treatment is short ( only once, and lasting 10min). Other trials, the position of treatment is carotid, where the flow speed of blood is quick, and the time of treatment is long. The last, at present, its not confirmed to the effect of RM. It had reported that RM has no effect apparently.We found that RM is effective on the expression of Bcl - 2 and Bax. The effect is apparently.
