Ihibitory Effects Of Extractive Liquid Warter From Centipede On The Proliferation Of Glioma In Vitro

Objective: To study the antineoplastic activity in vitro andits mechanism of Chinese medicinal Sclopendra subspinpesmutilans koch extract with water.To produce the scientificevidence for developing and manufacturing new antitumordrugs.Methods: The suppressive effect of Sclopendra subspinpesmutilans koch extract with water on the proliferation of C6tumor cells in vitro using cell culture technique and MTTcolorimetric;The C6 tumor cells PCNA and the tumor cellproliferation phases was studied by fluorescent taining;Theapoptosis mechanism of C6 tumor cells was studied by DNAagarose gel electrophoresis technique and fluorescent taining.Results: 1. Sclopendra subspinpes mutilans koch extractwith water inhibited markedly the proliferation of C6 tumorcells in vitro and there was better dose-dependentrelationship.The effect of the tumor cells inhibitory wasenhanced with the medicine dose.2. Proliferating cell nuclear antigen of the C6 tumor cellswere deregulated by Sclopendra subspinpes mutilans kochextract with water, which shows Sclopendra subspinpesmutilans koch extract with water could inhibit the proliferatingactivety of the C6 tumor cells.3. The C6 tumor cells after treatment with Sclopendrasubspinpes mutilans koch extract with water, the cells inG0/G1 phase were blocked and increased remarkably.But thecells in S phase were decrease remarkably afer treatment withSclopendra subspinpes mutilans koch extract with water.TheG2/M phase in tumor cells were not changed clearly.4. The DNA agarose gel electrophoresis technique resultsafter treatment of the Sclopendra subspinpes mutilans kochextract with water shows there were not characteristic "DNAladder"pattern.5. Apoptosis DNA after treatment with the Sclopendrasubspinpes mutilans koch extract with water checked up byfluorescent taining show that there was no obviously differencecompared with negative comparison.Conclusion: 1. Sclopendra subspinpes mutilans kochextract with water inhibited markedly the proliferation of C6tumor cells in vitro and there was better dose-dependentrelationship.2. Proliferating cell nuclear antigen of the C6 tumor cellswere deregulated with treatment of Sclopendra subspinpesmutilans koch extract with water, so it can inhibit tumor cellsproliferating activity and inhibit the growth ability.3. Sclopendra subspinpes mutilans koch extract with waterinhibit the tumor cells proliferating activity by inhibit the cellcycle mechanism.
