| Objective To investigate the etiopathogenisis of hypospadias through the establishment of congenital hypospadias model in mice and the stimulation of epidermal growth factor(EGF) in mice phallus with hypospadias using human chorionic gonadotrophin(HCG). Methods To select 144 mice (Kunming species), from the 15th day of their pregnancy, received gavage of flutamide for 5 consecutive days in a dosage of 50mg·kg-1·d-1,100mg·kg-1·d-1,200mg·kg-1·d-1,300mg·kg-1·d-1 or 400mg·kg-1·d-1; after withdrawal,the pregnant mice ran a spontaneous labour; from on the 21st day after birth the male whelps were observed on the morphological change of their external genital processes.Congenital hypospadias in mice(Kunming space) were established.To select 20 mice with normal and 20 mice with hypospadias from 3 weeks of postnatal weaning and to detect concentration of EGF in mice phallus using ELISA respectively.To select 50 normal mice with normal sodium of intraperitoneal injection was control group.To select 50 mice with hypospadias,10 mice of the total with normal sodium of intraperitoneal injection,40 mice of the total to 4 group with HCG of intraperitoneal injection for 7 consecutive days in a dosage of 50IU,100IU,150IU or 200IU respective. To detect concentration of EGF in mice phallus and serum with control group and test group using ELISA respectively. Results Group with 300 mg·kg-1·d-1 showed their external genitals were very much similar to the female mice's,with pubes around scrotum,testes unprotruded and cryptorchid; deformity rate up to 83.3%; group with 400 mg·kg-1·d-1 showed their urethral meatus was present on the ventral side of the penis, the penis was curved and the prepuce covered the dorsal side of the penis as the so-called skid hood, and even a vagina-like orifice formed in the ventral, deformity rate up to 94.4%.To detect concentration of EGF in mice phallus and serum with control group and test group using ELISA respectively.The total concentration of EGF in mice phallus with hypospadias was lower than these of control group (P<0.05). It was distinction between with HCG of intraperitoneal injection in a dosage of 150IU,200IU group and with normal sodium of intraperitoneal injection of mice with hypospadias group and with HCG of intraperitoneal injection in a dosage of 50IU,100IU group(P<0.05).It was not distinction between normal group(P>0.05).The total concentration of EGF in mice serum was not distinction with each group . Conclusions Congenital hypospadias in mice(Kunming space) were established using female mice (Kunming species), from the 15th day of their pregnancy, given a flutamide gavage in a dosage of 300 mg·kg-1·d-1 or 400 mg·kg-1·d-1 for consecutive 5 days. The nonsteroidal antiandrogen may be related to the aetiology of hypospadias. The total concentration of EGF in mice phallus with hypospadias is decreased,which may be related to the genesis of hypospadias.Exogenous of HCG could stimulate the increase of concentration of EGF in mice phallus with hypospadias in a dosage of 150IU or 200IU . |
