OBJECTIVE To study the pathogenic property and the molecular characterization of influenza viruses isolated from Qingdao during 2004-2005 in order to supply the basis on which the prevention and treatment of the influenza will be done.METHODS In this experiment ,196 clinical specimens collected from patient were propagated in MDCK cells (M) and 10-day-old embryonated hen'eggs (E) simultaneity, 12 was isolated from the first culture, and the results of hemagglutination test was nagetive were grown for the secondly time and another 4 were isolated in this method ,the isolated rate is 8.16%,and then Through the antigenic analysis, 4 H3N2 influenza virus , 2 H1N1 influenza virus and 5 influenza B virus were grown from embryonated hen'eggs , 2 H3N2 influenza virus , 1 H1N1 influenza virus and 2 B influenza virus were isolated in MDCK cells .The virus RNAs were extracted by trizol reagent and the RNA segment were amplified by reverse transcription-polymerase chain reaction (RT-PCR), then sequenced. The nucleotide sequences on HA protein molecular of A influenza and NP protein molecular of B influenza compared with those of predominated strain of influenza virus in china during the 1998-2004 virus respectively, and study on their Gene variation .RESULTS The 3 H1N1 influenza were high homology With A/New Caledonia/20/99(H1N1) ,the 6 H3N2 influenza were high homology With A/Sydney/5/97(H3N2) and the 7 B influenza were high homology with B/Shanghai/361/2002.CONCLUSIONS The result demonstrated that the epidemiologic characters of the Qingdao area in 2004-2005, and offer the import reference value for the vaccine recipe .
