Cervical cord compressive injury (CCCI) is a common disease which is caused mainly by degeneration of cervical vertabra and trauma. The accident of CCCI has increased recently, and it is difficult to recovery for the reason of secondary injury of nerve tissues. To treat CCCI patients, we must protect the neurons, prevent and alleviate SCCI, promote regeneration of neurons.Operation and non-operation are the two main treatment for CCCI. For the complex technique, patients with CCCI have to take some risks to have operation, and traditional Chinese medicine of non-operation has played an important role in some degree in the treatment for CCCI. Danshen Shujing pill has good clinical effects on CCCI, in 2003 it was funded by state Traditional Chinese Medicine Administration to study the effects of Danshen Shujing pill on regeneration of SCCI spinal cord, and there were 5 experiments as following in the project to study mechanism and effects of DSW treating CCCI.Aim:1.To study effects of DSW on general symptom and pathological changes of compressed cervical cord in CCCI rats; 2.To study the effects of DSW on apoptosis related gene in compressed cord in CCCI rats;3.To study the effects of DSW on reactive hyperplasia of neurogliainduced by compression in cervical cord in CCCI rats;4.To study the effects of DSW on regeneration of nerve fiber incervical cord in CCCI rats;5.To study the effects of DSW on the free redical in compressedcervical cord in CCCI rats.Material and methods:90 Wistar rats were used, 180g± 20g,cervical was compressed by a piece of silica gel at C4-C6, then the rats were divided into 7 groups, high-dose group, middle-dose group, low-dose group, JFK group, Model group, and JSH group, rats were feed after operation one week for one month with DSW (0.6g/ml, lml/lOOg, Qd), DSW (0.3g/ml, lml/lOOg, Qd), DSW (0.15g/ml, lml/lOOg, Qd), JFK (0.17g/ml, lml/lOOg, Qd), saline (lml/lOOg, Qd),and saline (lml/lOOg, Qd) respectively, and 8 rats of Model group were killed at 8th day after operation.After feeding drugs 30 days, the rats were narcosised by injecting 0.4% soluble pentobarbitone into abdominal cavity, then were killed by perfusing 4% polyformaldehyde solution in the aortic through the left cardial ventricle, the compressed cervical cord were removed for paraffin section, and HE staining, Weil's staining, Nissl staining, Holmes's methods and immunohistochemistry staining methods were used to observe pathological change, myelined fiber, perikaryon, bcl-2protein, Bax protein, neurobibril, Glial fibrillary acidic protein (GFAP), Growth-associated protein-43 and neurofilament,then normal and degeneral neuron were analysized with 3Y image analysis instrument. Blood was removed from left ventrical and injected into the tube with 3.8% citric acid rated with blood by 1:10, lml for SOD, lml for MDA. and used to measure the level of SOD and MDA in blood in CCCI rats.Results:1 Experiment study on effects of DSW on pathological changes of compressed cervical cord1.1 Areas of injured cervical cord treated by drugs were smaller than those of Model group, area of injured cord treated by DSW is located under the silica piece, while area of Model group became bigger than silica piece for the reason of responsive hyperplasia of fibrin connective tissue.1.2Normal neurons of DSW treatment group were more than those of Model group, in JFK and DSW group, the result was similar, but not as significant as in DSW treatment group.2 experimental study on the effects of DSW on apoptosis related gene in compressed cord in CCCI rats2.1 Bcl-2 positive cells of DSW (high- and middle-dose group) were more than those of JSH, JFK and Model group (P<0.001).2.2 OD of bcl-2 expression of DSW (low- and middle-dose) weremore than those of JSH(P<0.05), JFK (PO.Ol)group, OD DSWgroup were more than those of Model group(P<0.001).2.3 Bax positive cells of JFK group were more than those of (low- andmiddle-dose) and JSH group (PO.01), Bax cells of low-dose DSWgroup were less than those of other groups besides JSH group (P<0.05),and Bax cells of DSW low-dose group and JSH group were less thanthose of Model group(P<0.01).2.4OD of Bax expression of high-dose DSW group were more thanmiddle-dose DSW, Model and JSH groups(P<0.01).3 Experimental study on the effects of DSW on responsive hyperplasia of glia in compressive cervical cord in CCCI rats3.1GFAP positive cells in JSH group were less than those in the othergroups (P<0.05); 3.2GFAP positive cells in JSH and DSW groups wereless than those in Model groups(P<0.05);3.3There was no significant difference between the treating groups,treating and JSH groups;3.4Expression of GFAP in three DSW groups was different;3.5OD of GFAP expression in middle-dose DSW and Model groupwas more than high-dose DSW, JFK groups(P<0.05);3.6There was no significant difference between normal group and theother groups.4 Experimental study on the effects of DSW on regeneration of nerve fiber in cervical cord in CCCI rats4.10D of GAP-43 expression in low-dose DSW group was the highest,that of JSH group was the lowest, there was significant differencebetween groups(P<0.01);4.2There was no significant difference between high- and middle-doseDSW and JFK groups;4.3OD of NF expression in low-dose DSW group was higher than thatin middle-dose DSW and M group(P<0.05).5 Experimental study on the effects of DSW on the free redical in compressed cervical cord in CCCI rats5.1 SOD in JSH group was higher than that in middle-dose DSW, JFK, and Model groups(P<0.05);5.2 SOD in low-dose DSW group was higher than that in middle-dose DSW, JFK and M groups(P<0.05);5.3 There was no significant difference between low-dose DSW and JSH groups;5.4 MDA in Model group was higher than that in JSH groups(P<0.05);5.5 There was no significant difference between DSW and JFK groups.Conclusion:1 Experiment study on effects of DSW on pathological changes of compressed cervical cord1.1 DSW can alleviate SCCI, protect neurons, reduce nervous fiber demyelination induced by injure, promote recovery of degenerative neurons.1.2 JFK can not protect neurons as significantly as DSW.2 Experimental study on the effects of DSW on apoptosis related gene in compressed cord in CCCI ratsDSW can protect neurons of compressive areas by regulating apoptosis related gene expression.3 Experimental study on the effects of DSW on responsive hyperplasia of glia in compressive cervical cord in CCCI rats3.1 Compression can induce reactive hyperplasia of astrocyte; 3.2DSW can regulate the hyperplasia of astrocyte to provide a favored microenvironment for fibroconnective tissue recovery;3.3 JFK had no significant effects on hyperplasia of astrocyte.4 Experimental study on the effects of DSW on regeneration of nerve fiber in cervical cord in CCCI rats4.1 After treatment with DSW, there were regenerative nerve fiber incompressive area of cervical cord;4.2DSW can promote recovery and regeneration of injured nerve fiberin compressive area in cervical cord;4.3 JFK did not have as significant effects as DSW.5 Experimental study on the effects of DSW on the free redical in compressed cervical cord in CCCI rats5.1 DSW (low-dose) can remove the extra free radical in blood in...
