PrefaceAcute ischemia reperfusion injury is a patho - physiology phenomena that usually occurred after the treatment of acute myocardial infarction, such as thrombolysis in artery and PCI. Inland and oversea scholars are trying to understand and solve the problem. Recent reseaches show that heat shock protein can protect cardiac muscle against ischemia reperfusion injury. So how to increase the expression of heat shock protein is key point question that the scholars want to solve all along. Dopamine is the drug that is usually used to treat acute myocardial infarction. It can distend the coronary artery and increase cardial output to protect cardiac muscle. This study was designed to observe the relation between the expession of heat shock protein and the myocardial ischemia time, and we want to understand the influence of dopamine on heat shock protein 70 expression in different ischemic time in rat heart.Methods and MaterialsAnimals and materialsAdult wistar rats weighing 270il5g, supplied by the experimental animal center of China Medical University. Dopamine hydrochloride injection, supplied by Shang Hai Harvest pharmaceutical co. Ltd. HSP70 IgG was bought from SANTA CRUZ in America.Animal groups and model preparationRats were divided into twelve groups at random: No. 1 group to No. 7 giveSodium Chloride by intravenous injection and ligate the left anterior descending artery for 0min, 15min, 30mm, 45min, 60min, 90min respectively. No. 8 group to No. 12 groug give Dopamine hydrochloride injection by intravenous injection and ligate the left anterior descending artery for Omin, 15min, 30min, 45min, 60min, 90min respectively.Model of acute myocardial infarction was induced by temporary ligation of the left anterior coronary artery using a standardized surgical procedure. Under 20%urethane (0.5ml/0. lkg,ip) anesthesia condition, the rats were intubated and artificially ventilated, making an ECG before the operation. Then open the thorax, cut the third and forth ribs, ligate the left anterior descending artery, at the same time making an ECG once more to make sure AMI occur. After a while, undo the blood vessel to recover blood supply of the cardiac muscle for five hours. Then removal the heart, cut the cardiac muscle where was supply blood by LAD.Sample and parameters measurementsThe tissue is measured by means of western blot, and get the gray scale fig with Fluochim V2.0 software.Statistical analysisAnalysis was performed with SPSS statistical software 11.0. T test was used to analyse the data, a difference was considered significant at p <0.05.ResultsWestern blot show the groups of giving Dopamine hydrochloride injection and giving Sodium Chloride have distinct difference except for the No. 1 and No. 7 group.Analysis by SPSS T - test show that No. 2 to No. 6 and No. 8 to No. 12 have distinct difference p <0.05. No. 1 and No. 7 group have not distinct difference p >0.05.
