Objective: early in 1996,Welburn had found the hyper-insulinemia and low insulin sensitivity index in hyperten-sion.The disord of glycometabolism existed in nearly 50%hypertension patient,and it appeared as an abaissement of sugartolerance,insulin resistance and hyperinsulinemia.The endo-thelial cell functional disturbance played as an inherent phenom-enon in insulin resistance,and the disorder of NO(endothelialcell relax agent) might play an important role in the endothelialcell functional disturbance and insulin resistance.many re-searches had found that insulin resistance could depress theactivity of Na-+K-+ATPase in hypertension patient's red bloodcell . But the researches about how it effected the Na-+K-+ATPase of kidney were still not more.The kidney was animportant organ for the balance of water and electrolytes,butthe changes of the Na-+K-+ATPase's activity could cause thebalance's disorder.So we carried out this test to search the effectof insulin resistance on the activity of the NO,NOS ,Na-+K-+ATPase and Ca-2+Punp in spontaneously hypertensiverat's kidney.Methods: Eight-week-old male SH rats (n=16) were rand-omized in control group (Group C,n=8) and fructose-fed group(Group F,n=8). At the first of test, we measured the bodyweight,blood press,plasma glucose and insulin of the rats.After 8 weeks we execute the rats.When executing the rat,wemeasured its body weight,blood press,plasma glucose,insulinand glucose tolerance determination,also we examin -ated theactivity of the NO,NOS,Na-+K-+ATPase and Ca-2+Punp in rat'skidney.Results: 1. The change in body weight was not notablein two groups at the end of 8 weeks after treatment(P>0.05).Atthe end of 8 weeks after treatment,Fructose-fed group had ahigher systolic blood pressure (SBP) than control group (P<0.01)although both two groups had higher SBP than before.2. FBG,FINS,and ISI level : Before the test,FBG,FINS,and ISI level were not difference in two groups(P>0.05).Butafter the test,FINS was higher in Fructose-fed group comparedwith control group(P<0.05),and ISI was lower in fructose-fedgroup compared with control group(P<0.05). Before and afterthe test ,FBG,FINS,and ISI level were not difference incontrol group(P>0.05). But before and after the test ,the FINSand ISI level were different in Fructose-fed group(P>0.005).3. Oral Glucose tolerance test : In test, we found that theblood sugar level and the area of blood sugar at 30 minutes,60minutes and 120 minutes were higher than control group.Andthe highest blood sugar level appear at 30 minutes. The activity ofNOS in Fructose-fed group was 0.27U.mg. prot-1, and theactivity of NOS in control group was 0.22U.mg. prot-1,they hadno significant difference in statistics(P>0.05).5.the activity of Na-+K-+ATPase and Ca-2+Punp in rat'skidney: the activity of Na-+K-+ATPase in Fructose-fed groupwas 16.52 umol.pi.mg.prot-1.h-1, and the activity of Na-+K-+ATPase in control group was 12.93 umol.pi.mg. prot-1.h-1,Theyhad significant difference in statistics(P<0.05). The activity ofCa-2+Punp in Fructose-fed group was 4.41 umol.pi.mg.prot-1.h-1,and the activity of Ca2+-Punp in control group was 4.45umol.pi.mg.prot-1.h-1, they had no significant difference instatistics(P>0.05).Conclusions: The activity of NOS in Fructose-fed grouphad an increase tendency compared to the control group. It cluedthat the composition of the NO was active. we supposed thehigh concentratial insulin might irritate glomerular capillaryendotheliocyte to release more NO. But the level of NO inFructose-fed group was lower than that in control group. It cluedthat the expenditure of the NO was increase. We supposed thatthe oxyradical and glycosylation dead end product would causethe additional expenditure of the NO. The activity of Na+-K+-ATPase in Fructose-fed group had an notable increase...
