Objective: To determine the protective effect of NGF on hypoxic-ischemic(HI) neuronal cells of neonatal brain in vitro.Methods: â‘ HI model of neuronal cells of neonatal brain in vitro was induced by the deprivation of oxygen and glucose according to the improved Goldberg's method. â‘¡In order to find out the optimal concentration, MTT method was applied to measuring the vitality of neuronal cells treated with NGF in different concentration of NGF.The result was compared with the vitality of neuronal cells in the normal group,and in the control (HI) one at different time points(0,24hours after HI). â‘¢Neuronal cells in vitro were divided into 3 groups: the normal ,the control(HI), and the therapeutic group. The morphologic changes of neuronal cells were observed by light and electronic microscopes. The expression value of Cyt-C and AIF mRNA of different group were determined by immunocytochemistry and RT-PCR respectively.Results: â‘ The value of MTT in HI group was significantly lower than that in the normal group and the best therapeautic one. According to the value of MTT, 80ng/ml of NGF is the optimal concentration. â‘¡Compared with that of normal group and the therapeutic one, the value of Cyt-C and AIF mRNA of the control was significantly higher (P<0.05). There was no significant difference between the normal group and the therapeutic one. â‘¢Light and electron-microscopic observation showed that the apoptosis and the abnormal morphologic changes of neuronal cells were obvious in the control group, compared with those in the therapeautic group.â‘£NGF which was used at 24 hours after HI was less effective than that at 0 hour after HI.Conclusion: In vitro, HI model of neuronal cells of neonatal brain can simulate HI brain damage (HIBD). Exogenous NGF (80ng/ml) has the best protective effect on HI neuronal cells.The earlier NGF was used the better the result was.
