Objective To establish the orthotopic liver transplantation (OLT) in the rats from which we could understand mechanism and importance of graft rejection and tolerance in liver transplantation. Methods Two-cuff (portal vein and infrahepatic vena cava) technique was applied for the production of this aninmal model in Wistar rats and SD rats. Compared with the survival rate, complication and pathologic damage was observed in thes two groups between Wistar-Wistar and SD-wistar rats after liver transplantation. Results Average total time for operation was 105±10 minutes, the anhepatic of period on average was 19±2 minutes, the model successful rate was 83. 3% achieved in the experimental study. The survival time on average after operation was 5. 1±0. 3days, and the 5-day survival rate was 58% (10/17) . There was no immunopathology damage was during the first week after OLT procedure in Wistar-Wistar group. While, in SD-Wistar group the survival time was 3. 8±0.2 days, and the 5-day survival rate was 37. 5% (3/8) . The pathology damage could be found since 5-day after performed OLT. Conclusion Two-cuff technique was relatively and highly reproducible, which may provide a practical and stable experimental model for the study on liver tansplantation. Xenotransplantation may increase the degree of the damage in the receptors, and reduce theirs survival time, based on our OLT model experience.
