He also asserted that pituitary gland and adrenal gland play animportant role in general adaptation syndrome. Mason stated that stress is an Psychologicconcept rather than a Physiologic concept on the basis of classic stress concept. Nowadaysresearchers pay more attention to Psychologic stress .Moreover ,some researcher identifystress with psychologic stress.Stress calls neuroendocrine reaction of organism andgeneral physical reaction. Especially in metabolism , circulatory system, digestive organ,and immune system. The purpose of this study is to investigate some changes in digestiveorgan and immune system of rats under various psychologic stress patterns byimmunofluorescence detection of gastric H+,K+-ATPase and radio-immunity assay of seruminterleukin -2 level.Section One Immunofluorescence detection of gastric parietal cell undervarious psychological stress conditionsObjecetive: to investigate the relations of structure and function of gastric parietal cell undervarious psychological stress conditions by immunofluorescence detectionMethods: the experimental models of stress ulcer induced by various psychologicalstressors includes water restraint stress group, noise stress group and light-shock groupwere performed according to literatures previously in SD rats respectively.. The extentof gastric mucosal lesion was evaluated grossly and histologically in each group to some toGuth standard or Nils Lambecht standard .the pH value of gastric juice was measured by thepH/mV meter pH. The stomach was processed for immunofluorescence detection. BothImmunostaining of β subunit of H+,K+-ATPase and double staining for mitochondriaand αsubunit of H+,K+-ATPase were performed .The mucosa were examined and recordedby conventional microscopy and confocal laser scaning microscope (CLSM). We probed therelation of structure and function later. Results: 1. Gastric mucosal lesion: there was nolesion in control . there was a little line or lineal erosion or hemorrhage in light-shock group.The gastric mucosal ulcer index (UI) was higher in light-shock group (10.03±0.33) than incontrol (p<0.01). Some mucosal shedding and necrosis were observed under LM. Alittle bloodstain and some scattered line erosions and hemorrhage were observed in noisestress group . The UI (10.5±1.29) was higher in this group than in the control (p<0.01), andlots of necrosis, shedding and confusion were observed in water restraint stress groupgroup, more bloodstain and scattered spot or lineal erosions, hemorrhage and ulcer wereobserved. The UI (37.25±2.22) was much higher (p<0.01) in this group than in thecontrol(p<0.01), and in the noise stress group (p<0.01) or in the light-shock group. Andmore necrosis, shedding, hemorrhage, interruption and crateriform ulcer were observed..2.Gastric fluid pH value: The pH value was 2.59±0.21 in control and 2.19±0.07 inlight-shock group with statistical significant(p<0.05). The pH value of 2.22±0.05 in noisestress group .was lower than control(p<0.05) and 1.28±0.06 in WRS group muchlower than in other groups(p<0.01). 3. Quantitive analysis of the numbers of parietal cells :the fluorescence of parietal cells showed a reticular, diffuse, or mixed pattern incytoplasm .The reticular pattern corresponds to developed intracellular canaliculi inthe active stage of acid srcretion and the diffuse pattern to numerous tubulovesicles in theresting stage ..Quantitatively , 42.5±...
