Background: Viral myocarditis (VMC) is a well-known disease associated with characteristic lesions, such as necrosis of myofibers, inflammatory cell infiltration and fibosis. At precent, incidence of VMC is gradually rising and VMC has become a common disease. But so far, modem medicinal science has neither effective treatment to it, nor radical cure drugs. Traditional Chinese Medicine (TCM) has an evident advantage of preventing and treating VMC. Yixinyin is created by Professor Wei pinkang in terms of TCM theories and rich experience in clinic for many years.Objective: An experiment was done to research the effects of Yixinyin in experimental murine coxcackie B3 viral myocarditis in order to assess the efficiency of Yixinyin in treating VMC and develop a good medicine in Chinese herbs for clinical application.Methods: There are two parts in this study, in vivo experiment and in vitro experiment.The beating myocardial cells were obtained from the ventricular myocardium of newborn Sparaque-Dawley (SD) rats and incubated for 48 hours and inoculated with Coxsackie B3 virus as experimental model of viral myocarditis. With Yixinyin-contained serum added, the beating rate and survival rate of myocardial cells, and the activity of lactate dehydrogenase were measured at the lstt 3rd and 5th day after infection. The experiment is carried out by dividing into five groups.In the experiment, all the 125 Balb/c mice were randomly divided into six groups, namely, the normal control group, the pathologic control group, the Astragalus group, the high dose Yixinyin group, the middle dose group, and the low dose group. The viral myocarditis model was established by injecting CVB3 (0.2ml/per mouse) into abdominal cavity in Balb/c mice. Yixinyin groups fed with 10g/kg, 5g/kg and 2.5g/kg per day respectively. In each group, 6 mice were killed on the 7th day, and the rest on the 14th day. We determined SOD and MDA at the 7th and 14th day. The expression levels of perform and granzyme B mRNA in myocardium were measured by a semi-quantatative reverse transcription polymerase chain reaction (RT-PCR)procedure at the 7th and 14th day, while pathologic examinations of the hearts were performed.Results: Serum contained Yixinyin significantly alleviate the pathogenic changes of myocardial cells , improve percent of cell beating, reduce the activity of lactate dehydrogenasei increase the survival rate of myocardial cells infected with Coxsackie 83 virus. Symptoms, such as pilo-erection, lassitude, loss of body weight and loose stool, occurred on 2nd day. The death of mice occurred on 3rd day, reached climax from 5th day to 13th day. By 14th day, the number of death in all groups is 0,12, 6,6, 4, 2; Compared with the pathologic control group, SOD activities were increased and the level of MDA decreased in the middle and high dose Yixinyin group. The Yixinyin could significantly reduce the perform and granzyme B mRNA expression, it also could significantly alleviate the pathogenic and ultrastructural damage of myocardial cells.Conclusion: 1. Serum contained Yixinyin has significant protective effects on myocardial cells infected with Coxsackie B3 virus in SD rats (P< 0.05-0.01).2. Yixinyin is efficacious in reducing mouse death rate, improving general conditions and pathological and ultrastructural damage of myocadium for Balb/c mice.3. Yixinyin acts as an effective free radical scavenger and anti-lipid peroxidation drug (p<0.01). 4.Yixinyin has significantly effect in antagonizing immune injury for Balb/c mice myocardial cells infected with Coxsackie B3 virus (p<0.01), the mechanism of its immunosuppression could be through lowering the perform and granzyme B mRNA expression.At the end, combining the theories of TCM with modern researches, the thesis sets forth the foundations of Yixinyin'constructing formulate and using medicine, we consider Yixinyin as a important role in invigorating Qi and Yin, clearing heat and toxic material.
