Objective: Using a blood-brain barrier (BBB) model in vitro,we observe the effect of Borneol to the BBB,research the pathway which Borneol increases other substances through the BBB.And we would explain the "Kai Qiao "mechanism of Borneol. Method: Using the technique of brain miscrovessle endothelial cells (BMEC) and astrocytes (AC) co-culture,we established a BBB model in vitro.We used the transendothelial electrical resistance (TEER),mdrla mRNA act as the evaluating criterion,to observe the effect of Borneol to the BBB.And we determined the amount of Salb by HPLC-MS,to observe the effect that Borneol increased Salb through the BBB. Result: 1. Succeeding to culture BMEC, under the BMEC and AC co-culture condition ,we established a BBB model in vitro. 2. AC increased the Y -glutamyl transpeptidase (y -GT) and alkaline phosphatase (ALP) activity of the BBB, when co-cultured with BMEC. 3. AC increased the TEER and induced the tighe junctions (TJ) in BBB BMECs. 4. Borneol couldn't change the TEER in vitro BBB model in 60 minutes and 24 hours. 5. Borneol and Salb drastically decreased the mdrla mRNA expression in BMEC of the in vitro BBB model in common, whereas Borneol and Salb couldn't respectively.Conclusion: 1. Borneol couldn't induce the opening of tight junctions in BBB. 2. Borneol and Salb drastically decreased the mdrla mRNA expression in BMEC of the in vitro BBB model,which maybe was the one of mechanisms that Borneol increased other substances through the BBB and "KaiQiao".
