Objective: Oligodendrocytes develop from oligodendrocyte-type 2 astrocyte progenitor cells. To observe the effects of radiation on differentiation of O-2A, so as to provide basic data on pathogenesis of brain radiation injury.Methods: Differentiation of O-2A was observed after radiation in different dose(2Gy, 10Gy and 30Gy) by PO-P1 Sprague-Dawley Rat's mixed glial cells culture, induced differentiation and immunocytochemistry. After radiation the quantities of O-2A, morphological changes and the ratio of the differentiation were detected.Results: The mixed glial cells culture have two layers. A2B5-positived O-2A are the major cells on the superstratum and GFAP-positived type-1-astrocytes are the primary cells on the underlayer. After radiation the quantities of O-2A decreased significantly, Decreased shorter and thicker neuritis of oligodendrocytes and type-2-astrocytes were found. There were no significant changes of the ratio of the differentiated oligodendrocytes and type-2-astrocytes.Conclusion: The model of the mixed glial cells culture is propitious to know proliferation and differentiation of O-2A after radiation in vitro.
