Objective: Study the expression of tumor suppressor gene, DNA repair gene and telomerase component in acute leukemia, discuss their effects on the occurrence and development of leukemia and their clinical meanings .Methods: Collect marrow biopsy samples of 45 childhood acute leukemia(including 25 first diagnosed children and 20 remission children) , and 20 contrasts. Check the expression levels of PTENmRNA on paraffin-embedded sections of marrow biopsy samples by in situ hybridization, and check the expressions of hTERT and DNA-PKcs by immunohistochemistry.Compare the differences of three groups.Results: (1) PTENmRNA positive rates in groups of first diagnosed, remission and contrast are 60.0%,95.0% and 100%;loss expression rate of first diagnosed group is higher than the remission group and contrast group notably (P<0.05,P<0.01); (2) hTERT positive rates in groups of first diagnosed, remission and contrast are 84.0%,50.0% and 40.0%; positive rate of first diagnosed group is higher than the remission group and contrast group(P<0.05,P<0.01); (3) DNA-PKcs positive rates in groups of first diagnosed, remission and contrast are 72.0%, 100% and 100%,loss expression rate of first diagnosed group is higher than the remission group and contrast group(P<0.05); (4) there is relation between the expressions of PTENmRNA and DNA-PKcs in first diagnosedgroup(P<0.05);(5) patients with negative PTENmRNA expression have lower CR rate than positive ones(P<0.05); the CR rate of patients with positive hTERT expression is near negative ones; patients with negative DNA-PKcs expression have higher CR rate.but not notable(P>0.05).Conclusions: Childhood acute leukemia shows loss expression of PTENmRNA.high expression of hTERT and loss expression of DNA-PKcs, indicating that tumor suppressor gene, DNA repair gene and telomerase have an important role in the occurrence of childhood acute leukemia. Loss expression of PTENmRNA in our experiment may contributes to judging curative effect and estimate the prognosis.
