The Study Of Expression And Function Of Antigen Presenting Cell In Asthma

Objective To explore the changes of cells composition in broncholveolar lavage fluid ( BALF ) and peritoneal cavity irrigaion from mice sensitized and challenged with ovalgumin and function changes of peritoneal cavity macrophage and expression of spleen dentritic cells .This essay here intend to discuss some effections of antign on antigen presentation cells.Methods Mice were sensitized and challenged with ovalgumin to establish an asthmatic model . collect the BALF and peritoneal cavity irrigaion and observe the quantity changes of cells in different times .Macrophage la antigen expression of peritoneal cavity macrophage and the percentage of spleen DCs werw determined by flow cytometry .ELISA was directed to examine the levels of TNF- a and LT-C4 in peritoneal cavity macrophage cultrate supernatants .Results (l)After challenging, the sencond,fifth ,seventh day ,abdominal cavity macrophage (106/ml) ,total BALF cells (105/ml) and absolut number of alveolar macrophage ( 105/ml) of sensitized group is 7.59 1.27, 2.01 0.61 , 1.66 0.52; 19.43 6.37, 2.87 + 0.50, 1.84 0.40; 7.68 + 2.28, 1.40 0.45, 0.99 0.38. After challenging,the sencond,fifth, seventh day,abdominal cavity macrophage ( 106/ml) ,total BALF cells ( 1 05/ml ) and absolut number of alveolar macrophage ( 1 05/ml ) of sensitized and challenged group is 16.62 5. 56, 3.63 0.79, 2.55 0.72; 15.34 4.22, 5.03 + 0.56, 2.99 0.42; 17.10 2.04, 5.05 0.75, 3. 15 0.37. are more than control group 3.13 + 0.74 (106/ml), 0.65+0.17 (105/ml), 0.54 + 0.18 (105/ml) .(2)The percentage of la antigen expressing macrophage among sensitized challenged and control group is 44.09%, 59.28 %, 1.61%. As compared to control group, the peritoneal cavity macrophage la antigen expression of sensitized and challenged group was markedly increased(p<0.01), morever challenged group is more intensive than sensitized group(p<0.05). (3)The percentage of spleen DCs among sensitized challenged and control group is 1.84%^ 16.18%> 3.88 % The percentage of spleen DCs in sensitized group is higer than control group(p<0.01) . the contrast between challenged and control group has no difference(p>0.05). (4)The level of culture supernatants TNF- a (ng/ml) of abdominal cavity macrophage were 55.42 0.62 significantly higer than those of normal subjects5.73 1.83. (5)But the leverl of culture supernatants LT-C4 (ng/ml) of abdominal cavity macrophage is 7.37 0.41 did not differ clearly from those of nomal subjects6.97 0.54.Conclusions (1) The number of abdominal cavity macrophage absolut alveolar macrophageo in sensitized and challenged group are also markedly increased. (2) macrophage la antigen expression of sensitized group was markedly increased As compared to control group, so the peritoneal cavity macrophage antigen presentation function of sensitized an group was markedly increased, at the same time, challenged group increase more.(3) expressing of TNF- a were significantly higer than those of normal subjects, But the leverl LT-C4did not differ clearly from those of nomal subjects.(4) The expression of spleen DCs among sensitized is higer than control group,the contrast between challenged and control group has no difference.
