RNA interferance (RNAi) technology is a new method developed in recent years,which can restrict the gene expression . RNAi is the process that double-stranded RNA induces the homology-dependent degradation of cognate mRNA mediated by 21-23 nucleotide short interfering RNA ( siRNA ) . Here , we describe a simple DNA vector for efficient delivery of siRNA into lung cancer A549 cell utilizing the well-defined U6-RNA promoter, the vector can efficient delivered those temple into 21-23 nucleotide siRNA and those siRNA can induce the homology-dependent degradation of target gene . In this paper gene p53 was targeted by this vector .Objective: Through DNA vector-based RNA interferance technology to research silencing p53 gene in A549 cell.Methods: The DNA temple that was homology with p53 and was synthesized a special structure in vitro were transfected by Hposome into A549 cells. Proteins were extracted from cells and p53 levels were checked by Western blot.Results: Our results demonstrate efficient and specific knock-down of p53 in A549 cells using this DNA vector-based RNAi approach and indicate a promosing application of this method in lung cancer gene therapy .
