Objective: To determine the expression of P53, P21WAFI, MDM2, BAX and BCL-2 in esophageal squamous cell carcinoma and to investigate the correlativity of the genes. And to analyse the relationship between every protein expression and clinipathological characters. Method: The SP immunohistochemical method and two-step method were performed in tissue specimens of 43 cases of esophageal squamous cell carcinomas, 30 respectively cases of normal esophageal mucosas and dysplastic esophageal mucosas with antibody of those proteins. Result: The expression of every protein had the significant difference among esophageal squamous cell carcinoma, dysplastic and normal esophageal mucosa, of which P53 , MDM2 enhanced increasingly but conversely of P21WAFI, BAX. There exited the significant difference of MDM2 between every two kinds of tissue, but the significant difference of P53, P21WAF1, BAX were indicated between normal and carcinoma, dysplasia and carcinoma. The positive rate of MDM2 in lymph node transfer of 80% was higher than in non- lymph node transfer of 35.7%. The expression of P53, MDM2 increased but P21WAFI reduced with the stage rising in squamous cell carcinoma. We found the the difference of P53 and P21WAF1 in stage I and 2, I and 3 and MDM2 in stage I and III. There didn't found the correlation between P53, P21WAF1 and MDM2 but P53 and BAX. In addition, bcl-2 didn't express in all esophageal squamous cell carcinomas. Conclusion: 1.Abnormality of P5 3 , MDM2 , P2, BAX may be involved in occurrence and development of esophageal squamous cell carcinoma together.2. P53 contributes to esophageal squamous cell carcinoma, by reducing the expression of P21WAF1 and BAX. 3.0verexpression of MDM2 is related to infiltration and transfer and may suggested the bad prognosis. 4.The expression of P53 , P21, MDM2 have no correlationship. 5.Bcl-2 has few of roles in this carcinoma.
